| Recently, more and more attention has been given to the antitumor potential of venom. Among the various species of venom studied, cobra venom was found to be of the most potent antitumor activity and the membrane toxin is its major antitumor component It has been demonstrated that the antitumor spectrum of cobra venom is fairly broad Up to now, the published reports were restricted to the experimental study on the antitumor effect of raw venom. No clinical trial has been reported . Besides, its antitumor mechanism is still obscure. We investigated the direct antitumor activity and mechanism , and conducted a preliminary clinical trial of cobra venom against leukemia The results were as follows.1. MTT assay was used to detect the inhibitory effect of FNNAV on the proliferation of nine leukemia cell lines. The results indicated that FCFNNAV is toxic to tliese cell lines (IC50 were 0.0046- 4.40μg/ml) and the toxicity positively correlates with the doses applied (r were 0.66-0.99; p< 0.2-0.001 ).2. The toxicities of FNNAV to K562/S, K562/AO2 cells were similar. (IC50S were 1.86μg/ml and 2. 26μg/ml respectively, p>0. 05)3. Low concentration of FNNAV can reduce the IC50 of adriamycin (ADM) against K562/S and K562/AO2 cells (2.83μg/ml to 1. 48μg/ml and 196. 55μg/ml to 120. 98μg/ml respectively, both P<0.05), thus enhance the toxicity; and low concentration of ADM can strengthen the inhibitory effect of FNNAV against leukemia cells, indicating that ADM and FNNAV... |
