As an important chemical mediator and cellular signaling molecular, nitric oxide has been almost always attracting the attention of every researcher in the biomedical domain . During the past decade, an enormous number of studies have examined the modulating role of this unique gas molecule in every physiological and pathophysiological process. The relationship between nitric oxide and myocardial ischemia reperfusion injury is especially a hotspot of attention. Unfortunately, the change rule and role of NO in myocardial ischemia reperfusion injury are not clear by far because the half-life of free NO in physiologic solution is very transitory and direct measurement to it is very difficult. Our research is aimed to explore these questions by utilizing the advanced NO direct measurement method.With the widespread use of propofol in cardiac surgery and postoperative sedation management, the myocardial preservation induced by propofol is increasingly reported recently. Therefore, this study also investigated the relationship between NO and the myocardial preservation of propofol in order to perfect its cardioprotection mechanisms and to provide experimental clinic evidence to the theory of myocardial ischemia reperfusion injury.Part One The influence of ischemia reperfusion on myocardial endogenous NO system30 healthy adult SD rats were selected to fabricate the isolated heart preparation with Langendorff setting. After equilibration, the ISO-NO sensor was placed into the right atrial chamber closest to the coronary sinus in order to measure NO continuously in the coronary effluent during the ischemia reperfusion process.
