Investigation Of Esophageal Carcinoma Initiating Cell Isolation And Biological Behavior Characteristics

Object: In order to investigate a method of isolating esophageal carcinoma cell line(EC9706) initiating cell, preliminarily identifying the biological characteristics of it.Methods: EC9706 labeled with hoechst33342 was separated by flow cytometry into two groups: hoechst33342 negative/dim (side population, SP)and hoechst33342 positive (non-SP). The two groups were relabeled with hoechst33342 after cultivation in the same medium to identify the potentiality of self-renewal and multi-differentiation. The clonogenesis ability of the two groups was tested by soft agarose assay and nude mice xenograft model. The differences of ABCG2 expression, cell cycle, curve of growth and sensitivities to chemotherapeutic agents between the two groups was determined by RT-PCR, flow cytometry and MTT assay respectively.Results: The proportion of SP cell and non-SP cell was 4.5% and 95.5% respectively.
