Object:Obesity is an important disease which is harmful to the health of humankind. Obesity has close relationship with the occurrence of coronary heart disease, stroke, type 2 diabetes, hyperlipidemia and sleep apnea syndrome et al. The proliferation , differentiation of preadipocyte and the adipogenesis of adipocyte are the core of the development of obesity. Peroxisome proliferator activated receptor gamma (PPAR gamma) plays a key role in this progress. The 3T3-L1 cells stably transfeted by a luciferase expression plasmid which contains the PPAR gamma2 promoter was used as a tool to screening drugs. The aim of the this study is to find out throse which may influence the activity of the PPAR gamma2 promoter from the numerous natural compounds in the Chinese medical herbs, then to observe the effects of these drugs on weight, metabolism of glucose and lipid of SD rats.Methods:1. Cloning luciferase expression plasmid containing the human PPAR gamm2 promoter and establishing stable transfection 3T3-L1 cells line: The plasmid (pGL3- PPARγ21kb-Luc) which contains the human PPAR gamma2 gene promoter (-910~+140 bp) was used as model sequence to clone the 625bp (-615~+10bp) PPAR gamma2 promoter, the 625bp promoter was inserted into a luciferase reporter gene vector pGL3-Basic.
