Part One: Growth Inhibition of Human Brain Glioma Cell Lines SF767 and T98MG by Gleevec in VitroObjective To study growth inhibition of human brain glioma cell lines SF767 and T98MG by Gleevec in vitro,and try to disclose intrinsic molecular mechanism. Method The growth inhibition of SF767 and T98MG by Gleevec in vitro was evaluated with MTT assay and clonogenic growth assay.; Cell cycle distribution induced by Gleevec was analyzed by flow cytometry(FCM); Cells were divided into three groups: control, cells treated by Gleevec 8uM for 12hours and cells treated by Gleevec 8uM for 24hours;and the expressions of P21,Bax,Rad51mRNA were analyzed by immunofluorescene quantitative RT-PCR. Result Gleevec can inhibit the growth of SF767,T98MG in MTT assay and clonogenic growth assay in dose-dependent manner, and IC50 was about 8uM. Cell cycle was arrested in G1 phade . RT-PCR showed the upregulation of P21mRNA and downregulation of Rad51mRNA in SF767,T98MG treated by Gleevec. Conclusion: Gleevec showed inhibitory ability to SF767,T98MG in vitro ,and cell cycle arrest was the main celluar mechanism,and upregulation of P21mRNA and downregulation of Rad51mRNA maybe participated in molecular mechanism.
