Objective: To investigate the effect of γ-interferon on antineoplastic capacity of tamoxifen against breast cancer cells in vitro, the morphological changes,cell cycle and apoptosis of breast cancer cells(ER+ or ER-)affected by tamoxifen lonely or combined with interferon, estradiol were examined with microscopy, MTT assey,Flow cytometry, DNA gel electrophoresis and eletromicroscopy.Methods : Cell culture technique was used to culture human breast cancer cell line MCF-7 and MDA-MB-231 in vitro.After MCF-7 and MDA-MB-231 breast cancer cells were treated with tamoxifen, γ -interferon and estradiol alone or in combination for 12 24 36 48 60h, morphological changes were examined with microscopy, cell proliferation was evaluated by MTT assay, distribution of cell cycle was determined by flow cytometry, cell apoptosis was determined by DNA gel electrophoresis,FCM and eletromicroscopy.Resu1ts: MCF-7 and MDA-MB-231 cell lines were maintained in RPMI-1640 supplemented with 10% inactivated NBS.
