Objective: 1.To investigate whether tetramethylpyrazine inhibits growth of human retinal pigment epithelial(RPE) cell in vitro. 2. To study the expressing of hypoxia-inducible factor-1α(HIF-1α) protein and its correlated cell factor VEGF mRNA and VEGF protein induced by advanced glycation end products (AGEs) on retinal pigment epithelial (RPE) cells. 3. To study the effects of Tetramethylpraine(TMP) on the expressing of HIF-1αprotein , VEGF mRNA and VEGF protein induced by AGEs on RPE cells. TO study the molecular mechanism and offer helpful experiment data for the therapy of DR by tetramethylpyrazine.Method: 1. Human RPE cells were cultured by enzymatic dissociation methods. The purity of cultured RPE cells was assessed morphologically and confirmed by immunocytochemical staining for human cytokeratin. Subcultures between 3 and 8 (p3-8) were used in the study. 2.The tetrazolium dye-reduction assay (MTT) and cells counting method was used to determine the effect of tetramethylpyrazine on RPE cell survival and proliferation for 24 and 48 hours at eight concentrations(1, 10, 20, 40, 80, 160, 320, 640mg/L). The arrest rate of RPE cell proliferation by tetramethylpyrazine was calculated. 3.Different concentration of AGEs acted on RPE cells for the same time, the same concentration of AGEs acted on RPE cells for different time,...
