Objective: This study was aimed to investigate the pathogenesis of osteolysis and aseptic loosening of articular prosthesis, to clarify the possible role of RANKL-RANK-OPG system on the osteolysis in aseptic loosening of hip prosthesis, and to explore the effects of anti-inflammatory drugs (steroid, NSAIDS and Cox-2 inhibitor) on RANKL-RANK-OPG system pathway.Methods: This study was composed of two parts: Part 1- detecting of the expression of RANKL, RANK and OPG mRNA in the soft tissue around the failed hip arthroplasty. The penprosthetic soft tissue was obtained from 6 patients who suffer from osteolysis and aseptic loosening after total hip arthroplsty. Immunohistochemistry for RANKL and RANK in the penprosthetic soft tissue of revised hip arthroplasty were performed with the anti-RANKL monoclonal antibody and the anti-RANK monoclonal antibody.
