Objective To explore the effection of the cyclooxygenase-2 on the proliforation, metastasis, apoptosis of nasopharyngeal carcinoma. Methods 1. Immunohistochemical methods were performed to measure the level of C0X-2mRNA, COX-2, E-cad expression in collected samples of normal and immflame issues and human nasopharyngeal carcinoma issues. All the immmunohistochemistry results were measured by image analyze. 2. In vivo cultured CNE cells were treated with different concentration celecoxib, a selective COX-2 inhibitor. MTT, immunohistochemistry, electromicroscope , flow cytometryassay and Tunel were used to measure the proliferation, metastasis and apoptosis. Immunohistochemical methods were performed to measured the level of E-Cadin expression in CNE cells. The immmunohistochemistry results were measured by image analyze. 3. Human NPC transplanted subcutaneously in nude mice model was established, then , divided into 2 groups: control groups and celecoxib groups. All mice were sacrificed after 35 days, turmor volume and weight, COX-2,VEGF, MVD were detected. COX-2, VEGF and MVD were calculated by immunohistochemistry. Results 1.
