Hantaan virus (HTNV), the etiologic agent of hemorrhagic fever with renal syndrome (HFRS), is the prototype of the Hantavirus (HV) genus of the Bunyaviridae family. It has a single-stranded, negative-sense, tripartite RNA genome, whose segments are designated as large (L), medium (M) and small (S), which encode the viral RNA polymerase, envelope glycoproteins (G1, G2) and nucleocapsid protein (NP) respectively. The envelope glycoproteins can elicite neutralizing antibodies and protective cell-mediated immune response ; the nucleocapsid protein can only elicite protective cell-mediated immune response.In serological investigation, it has been reported that Hantavirus NP has strong antigenicity and immunogenicity. The antibodies against NP in patients with HFRS not only appear early, but have high titer. Most of these 100 more or less amino acids (aa) that are located at the amino (N) -terminus of HV nucleocapsid protein have hydrophilicity, and the N-terminal peptide can be recognized by sera of HFRS patients and many anti-NP monoclonal antibodies.
