| Osteogenic protein - 1(OP-1), or bone morphogenetic protein - 7 (BMP — 7) is a member of the transforming growth factor - beta superfami-ly of proteins and falls within the Dpp/Vg1 subgroup, being most closely related to bone morphogenetic protein -5,-6,-8(OP-2) . The cDNA was originally isolated from hippocampal and placental libraries using sequence information derived from peptides of bovine osteogenic protein preparations. The full length OP - 1 cDNA are all 1878bp. Open reading frame of OP - 1 with 1296bp encodes 431 amino acid residues, composed of signal peptide, pro - peptide, and mature peptide which is a highly conserved 7 cysteines transforming growth factor - beta domain in its C- tefminal and needs to be processed to gain its activities. Human OP—1, firstly, induces formation of bone, reparative dentine and periodontal regeneration in vivo. And It increases cell proliferation and collagen synthesis of osteoblasts in vitro. In addition, hOP — 1 stimulates increased expression of markers characteristic of osteoblast phenotype, namely alkaline phosphatase activity, osteocalcin synthesis, and PTH stimulated intracellular cAMP levels. Secondly, hOP - 1 may represent a novel class of neuroprotective agents. Thirdly, OP — 1 is a most important universal regulator of animal development.
