Objective:The research was designated to explore the change of cytokine and apoptosis-associated gene of experimental model rats of chronic pelvic inflammatory disease(CPID), and the mechanism of Yankening IV in treating CPID, which could provide evidence to the clinical application of YankeNing IV .Methods:Mixed bacterium was used to establish the CPID rats model. Sixty-seven female Wistar rats were randomly classified into the following different groups: the normal group, the simulative model group, the model group, the high dose of YankeNing IV group, the medium dose of YankeNing IV group, the low dose of YankeNing IV group and the control group of Chinese medicine. The histopathological changes of uterus was detected with the method of HE stain and the ultrastructural changes of uterus with electron microscope. The expressiom of Bax, NF-κB and ICAM-1 was detected with the method of immunohistochemistry and the expressiom of NF-κBmRNA and ICAM-1 mRNA in uterus was by RT-PCR. The content of TNF-a and IL-10 of serum was detected with the method of ELISA and the content of NO, NOS and SOD in serum was respectively detected with the method of nitrate reductase, chemical colorimetry and xanthine oxidase.Results:①Compared with the model group, the three YankeNing IV groups can obviously relieve hyperemia, edema and adherence, and accelerate the recovery of the tissues. The effect of the high dose of YankeNing IV group and the medium dose of YankeNing IV group were superior to the control group of Chinese medicine (P<0.05) ;②The YankeNing IV group can obviously decrease the expression of NF-κB and ICAM-1 mRNA. The effect of the high dose of YankeNing IV group was superior to the model group, the low dose of YankeNing IV group and the control group of Chinese medicine (p<0.05);③The YankeNing IV groups can obviously decrease NF-κB and ICAM-1 protein and increase the expression of Bax protein. The effect of the high dose of YankeNing IV group functioned significantly better than the model group, the low dose of YankeNing IV group and the control group of Chinese medicine (p<0.05);④Compared with the model group, the YankeNing IV groups can obviously decrease the contents of TNF-a ,IL-10, NO, NOS and SOD in serum. The high dose of YankeNing IV group functioned significantly better than the model group, the low dose of YankeNing IV group and the control group of Chinese medicine (p<0.05).Conclusions:①Mixed bacterium inoculated can effectively be used to establish the model rats with CPID;②The YankeNing IV can relieve hyperemia, edema and adherence, and accelerate the recovery of the tissues;③The YankeNing IV can decrease NF-κB and its downstream products (ICAM-1,TNF- a,L-10,NO and NOS);④The YankeNing IV can promote the expression of Bax, and thus promote apoptosis of inflammatory cell;⑤YankeNing IV can increase the activity of SOD, and accelerate clearning oxygen radical;⑥The YankeNing IV treats CPID by inhibiting NF-κB and the relevant inflammatory medium, promoting apoptosis of inflammatory cell and cleaning oxygen radical.The research has improved that the defined formula-Yankening IV was effective when treating the common gynecological disease-chronic pelvic inflammataotry disease(CPID).
