| Part One Induction of lupus nephritis models of intraperitoneal injection lipopolysaccharideObjective:To investigate the method to establish murine lupus nephritis models by intraperitoneal injection lipopolysaccharide (LPS). Methods: 80 mice were divided into four groups and were given NS or three different LPS doses (5mg*kg-1, 2.5mg*kg-1, 1.25mg*kg-1) respectively. Results: Compared with control group, the pathological change of kidney were appeared in 5mg*kg-1 and 2.5mg*kg-1 groups. The content of protein in 24 hours urine, the serum antinuclear antibody, and the deposition status of immune complex in nephritis of lupus were remarkably increased in 5mg*kg-1 group and 2.5mg*kg-1 groups than those in control group and 1.25mg*kg-1 group. The mortality of mice with LPS i.p in 5mg*kg-1 group was remarkably increased than that in other groups. Conclusion: The dose of LPS with 2.5mg*kg-1 i.p. injected can induce lower mortality than with 5mg*kg-1 in establishing SLE models and it can be used as an appropriate model for further research.Part Two Effects of combined treatment of traditional Chinese medicine and western medicine on Murine Lupus Nephritis ModelsObjective: To study the effects of combined treatment of traditional Chinese medicine and western medicine on Murine Lupus Nephritis Models. Methods:120 mice were made to murine lupus nephritis models with LPS by intraperitoneal injection. These mice were divided into six groups and were given distilled water, Qingrejiedubushen Decoction, prednisone, cyclophosphamide, Bushenqingrejiedu Decoction plus prednisone, and Qingrejiedubushen Decoction plus cyclophosphamide by intragastric administration (1 time per day) for 15 days respectively. Results: Compared with control group, the mortality, the content of protein in 24 hours urine, the serum antinuclear antibody, and the deposition status of immune complex in nephritis of lupus were remarkably decreased in all treatment groups, especially in combined treatment of traditional Chinese medicine and western medicine groups. Conclusion : Prednisone, cyclophosphamide, and Qingrejiedubushen Decoction all have therapeutic effects on Murine Lupus Nephritis Models, and the therapeutic effects of combination of traditional Chinese medicine and western medicine are better than others. |
