Objective To investigate expression rule of S100P by the real-time-PCR technology and impact on growth of pancreatic cancer cells in vivo and in vitro. To evaluate potentiality of S100P as a molecular marker of gene diagnosis and therapy. Materials and methods To compare the expression of S100P gene between 20 pancreatic cancer, 10 chronic pancreatitis and 10 normal pancreatic tissues, calculate the relative contents by RQ-PCR technology. Lentivirus with S100P gene was constructed, impact of S100P on pancreatic cancer cells was examined by MTT and FCM. Lentivirus with S100P-GFP fusion gene was constructed and pancreatic cancer cells with stable high expressions of S100P were filted. To establish implant pancreatic cancer model on nude mouse and to observe impact of S100P on growth of tumour . Results The relative contents of S100P in pancreatic cancer tissues were more than those of pancreatitis tissues. Two groups had significant differences. Normal pancreatic tissues almost had no expression .There was significant differences between negative and positive of lymphatic metastasis, and the similar phenomena was found between TNM stage I, II and those of stage III and IV. S100P gene had no notable impact on growth of pancreatic cancer cells in vivo.There was significant difference in volume of graft tumor between experimental and control (experiment and SW1990) group, at meanwhile, S100P mRNA determined high-expression in experiment graft tumor tissue comparing with control. Conclusion Expression up-regulation of S100P gene maybe an agent of pancreatic cancers.Perhaps S100P had important function on lymphatic or distant metastasis. S100P could be used as a molecular marker of gene diagnosis, gene therapy and prognosis predict..S100P gene was found to enhance the growth of pancreatic cancer cells significantly in vivo of nude mice, but to have no impact on the growth of pancreatic cancer cells in vitro.
