Objective: To study the effect of Yiqi Yangyin SanjiePrescription(abbreviated as YYSP) (contained in serum) on proliferation,apotosis, related gene PCNA and survivin of lung carcinoma cells from thelung cancer patient deficient in both Qi and Yin, and explore its mechanismagainst lung carcinoma.Methods: One case of lung carcinoma patient deficient in both Qi and Yinis selected and his separated cancer cells are cultured as subject. YYSPis administered by gastric garage to normal rats to produce the serumcontaining YYSP, which is used as the group with medicine. The rat serumwithout YYSP is taken as the control group. The effects of two differentserum on carcinoma cells are studied. 1) the effect of serum containingdifferent YYSP concentration on the proliferation of lung cancer cellsis detected by applying MTT, 2) the effect of serum containing YYSP onthe PCNA mRNA expression of lung cancer cells is detected by RT-PCR; 3)the effect of serum containing YYSP on the apotosis of lung cancer cellsis detected by annexin V/PI; 4) the effect of serum containing YYSP onthe expression of survivin mRNA is detected by RT-PCR. All data arestatistically dealt with by SPSS12 software.Results:1. Comparing with the control group, in the groups with medicine, YYSP concentration of 5%, 10% and 20% significantly inhibit theproliferation of lung carcinoma cells (P<0.05). The inhibiting effectof the serum YYSP concentration of 10% is obviously stronger than thatof the concentration of 5%(P<0.05), while the concentration of 20% doesnot show apparent difference from that of the concentration of 10%(P<0.05)2. Comparing with the control group, the expression of PCNAmRNA in thegroup with medicine decreases extremely significantly (P<0.01).3. Comparing with the control group, the rate of apotosis in the groupwith medicine increases extremely significantly (P<0.01).4. Comparing with the control group, the expression of survivin mRNA inthe group with medicine decreases significantly (P<0.05).Conclusions:1. Serum containing YYSP can obviously bring about the inhibition ofproliferation and induce apotosis of lung carcinoma cells from the lungcancer patient deficient in both Qi and Yin, which may be an importantmechanism in treating lung carcinoma.2. Serum containing YYSP can decrease the PCNA gene expression of lungcarcinoma cell, which may be the molecular mechanism of its inhibitingeffect on the proliferation of lung carcinoma cells.3. Serum containing YYSP can decrease the surviving gene expression oflung carcinoma cell, which may be the molecular mechanism of itsinducing effect on the apotosis of lung carcinoma cells.
