| Objective: Reperfusion injury is the main reason for early graft failure after lung transplantation. Chinese herbal medicine Xuebijing possesses some compounds which contain antioxidant properties and some other biological activities. We tested here the hypothesis that Xuebijing could ameliorate the lung reperfusion injury by vary mechanisms.Methods: Experiments were carried out on 40 adult rats of either sex. The animals were randomly divided into two groups of 20 animals each (LPD group and Xuebijing group). The lung ischemia reperfusion injury model was established as rabbit orthotropic pulmonary autograft transplantation model. Xuebijing was given in Xuebijing group. Reperfusion time was 2 hours. Left pulmonary vein blood and left lung were taken after 2 hours reperfusion. Pressure of Oxygen(PO2) in the Left Pulmonary Vein was measured. Apoptosis index (AI) was estimated by TUNEL assays. Lung wet/dry weight ratios(W/D) were estimated. Superoxide dismutase (SOD),malondialdehyde(M DA),myeloperoxidase (MPO) of lung tissue were detected. Lung specimens were obtained to observe the histological changes by microscopy and transmission electron microscopy.Results Edema and Inflammatory Cells emigration were more serious in the LPD group than that in xuebijing group in the pathology slice. Transmission electron microscopy examination showed more severe injuries in the LPD group than in the xuebijing group. The AI was significantly higher in LPD group(11.36±3.81) than in xuebijing group(7.95±2.68). The W/D was significantly lower in xuebijing group(5.69±0.23) than that in LPD group(6.78±0.31). PO2 of left pulmonary vein was significantly difference between the two groups (xuebijing group :58.9±9.3 mmHg,LPD group:45.3±8.7 mmHg, P<0.05). The SOD activity of Xuebijing group (0.7632±0.1067U/mg.pro) was significantly higher than those of LPD group(0.4254±0.0628 U/mg.pro, P<0.01)。The levels of M DA in lung tissue of Xuebijing group(0.1685±0.0511nmol/mg.pro ) were lower than those of LPD group(0.4398±0.1026 nmol/mg.pro ,P< 0.01).The levels of MPO in lung tissue of Xuebijing group(0.18±0.02?OD/min.g) were lower than those of LPD group(0.48±0.14?OD/min.g ,P< 0.01).The result suggested that Xuebijing could inhibited neutrophils activation and improved the anti—free radical ability of the lung. Xuebijing could improved the graft lung functiong by reducing lung edema and inflammatory after transplant surgery.Conclusion Xuebijing is an antioxidant, it can reduced the lung injury induced by oxygen radicals. Xuebijing can reduce edema and inflammatory of ischemia reperfusion lung, decrease apoptosis cell in the lung during ischemia reperfusion injury. We conclude that Xuebijing can attenuate lung ischemia reperfusion injury and protect the lung from graft failure. |
