objective:To observe the effect of Compound QiJie Cream (CQJC) in rat cutaneous incised wound healing and investigate the mechanism of accelerating the rat back wound healing.Methods:two full thickness dermal excision wound models with the diameter of 1.8cm were established on the back of 72 healthy male or femal Wistar rats, which were randomly divided into 3 groups:model group, Jing wanhong goup(Control goup), CQJC goup,24 rats per group. Jingwanhong as Positive compare medicine.The rats in each group were extemal applied with corresponding drug,0.1 g each wound surface and twice a day. Tissues were removed 3,7,10and14 days after surgery.The change of histomorphology was observed by the means of hematoxylin-eosin (HE) staining, and the fibroblasts in granulation tissues of the wound in each group were observed by PCNA staining. The CD31 staining was used to study the new blood capillaries in granulation tissues. The content of EGF, TGF-β1, Collagen-â… , Fn, Collagen—Ⅲand EGFR was determined by the use of streptavidin-perosidase immunohistochemistry. The lysozyme in wound tissue was tested by the means of streptavidin-perosidase, and the fibroblast cellular apoptosis in granulation tissues was studied by TUNEL method. Images of each group were analyzed by Image-Pro-Plus image analysis software. All counted data was expressed in the form of x±s, and processed by SPSS16.0 statistics software.Results: The wound healing rate, wound healing time, wound granulation tissue thickness, the scar area after 30days and other indicators of the CQJC were significantly heighten than Model group(P<0.05 or P<0.01) in each time points of observation; The new blood capillaries in the wound granulation tissues and the fibroblast cells of CQJC and Jing Wanhong group were statistically increased (P<0.01)than that of model group at day 3, day 7 and day 10 after surgery. The content of EGF, TGF-β1, Collagen-â… , Fn, Collagen—Ⅲand EGFR of the CQJC group was improved comparing to the model group and Jing Wanhong group. The content of wound lysozyme of CQJC group was obviously higher than other groups. When applied with CQJC at day 3 and day 7 after changing dressing, the wound cellular apoptosis was distinctly increased, and after that it turned to normal level gradually.Conclusion: The CQJC can promote wound healing in rat skin incision, reduce the scar size and improve the quality of wound healing. This can be explained that the CQJC may facilitate the proliferation of new capillaries and fibroblast cells in the early stage, as well as the growth of wound granulation tissue, to enhance the wound healing. The content of collagen protein can be improved by raising the amount of EGF, TGF-β1, Collagen-â… , Fn, Collagen—Ⅲand EGFR, to enhance the wound healing. Meanwhile, the CQJC can enhance the content of wound phagocyte lysozyme, induce the active apoptosis, and to be good for debridement of the wound in early stage. |
