Objective: The aims of this study are to assess the protective effect ofelectro-acupuncture "Xinshu" and "Jueyinshu" on myocardial ischemia reperfusion injuryin rats,and to explore mechanism of anti-apoptosis related factors for acupuncture"Xinshu" and "Jueyinshu" on myocardial ischemia reperfusion injury.Methods:All the wistar rats were randomly divided into10groups(n=8in eachgroup):(1)Sham-operation groupⅠ,Ⅱ,(2)Model groupⅠ,Ⅱ,(3)Treatment groupⅠ,Ⅱ,(4) Positive control groupⅠ,Ⅱ,(5) Negative control groupⅠ,Ⅱ. Series Ⅰweretreated for3days and series Ⅱ were treated for7days with electro-acupuncture.A comparative study on the apoptosis index(AI), were conducted The expressioncharacteristics of target gene Fas/FasL, high temperature requirement A2(HtrA2),5-lipoxygenase(5-LOX) and Cytochrome P450monooxygenase(CYP450) in myocardialtissue were achieved. The apoptosis related factors and acupuncture relevance wereanalyzed. The plasma cytokines TNF-α and ICAM-1were also measured by ELISA.Results:1. Compared with Sham-operation group, the expression ofpro-inflammatory factors TNF-α and ICAM-1, the apoptosis index(AI),and the apoptosisrelated factors i.e. Fas,FasL, HtrA2,CYP450were significantly upregulated in Modelgroup.2. Compared with Model group, the apoptosis index, and the apoptosis related factorsi.e. Fas, FasL, HtrA2, CYP450were significantly downregulated, whereas P38mitogen-activated protein kinase P38(P38MAPK) was increased in TreatmentgroupⅠ(treated3days). There were no significance on plasma profiles of TNF-α and ICAM-1.3. Compared with Model group, the plasma profiles of TNF-α and ICAM-1, AI, andthe apoptosis related factors Fas, FasL, HtrA2,CYP450were significantly downregulated,whereas P38mitogen-activated protein kinase P38(P38MAPK) was increased inTreatment group Ⅱ(treated7days).4. In the Treatment group, our findings indicated that the level of plasma cytokinesTNF-α and ICAM-1, and the apoptosis related factors Fas, HtrA2, CYP450in GroupsⅠ,which were treated3days, were lower than that of Groups Ⅱ.On the other hand, the AIand the expression of FasL, P38MAPK,5-LOX in GroupsⅠ were higher than that ofGroups Ⅱ, which were treated7days.Conclusion: In conclusion, these results clearly indicated electro-acupuncture"Xinshu" and "Jueyinshu" protects myocardial tissue in MIRI, due to altered expressionlevels of the apoptosis related factors and inflammatory factors; meanwhile increasing theexpression of P38MAPK. Our results also raise the possibility that the Treatment7daysGroup has better protective effect than Treatment3days Group at the early stage of theMIRI.
