| OBJECTIVEThe experiment was figured out that the influence of estrogen in the expression of P2X3and P2Y1receptors in the trigeminal ganglion in model of migraine rats.METHODSFirstly, we developed a rat model of nitroglycerin(NTG) induced to migraine. The ethology of rats in model group and control group was observed. Then P2X3and P2Y1receptors in the trigeminal ganglion of rats were measured by RT-PCR,Western-blot and Immunohistochemistry to figure out the expression of the receptors and the difference of them by gender. Secondly, we continue to discuss the role of estrogen effecting P2X3and P2Yj receptors in the trigeminal ganglion of migraine rats. Some female rats were divided randomly into shamoperated group(Sham), ovariectomized group(OVX) and estrogen replacement group(E2-R). The three groups were made models of migraine. Then the ethology of rats in all three groups were observed. Detect the expression of P2X3and P2Y1receptors by means of RT-PCR,Western-blot and Immunohistochemistry.RESULTSFirstly, the data of ethology was revealed, compared with control groups, rats in model groups had significant painful behavior (p<0.05) The expression of P2X3and P2Y1receptor in the trigeminal ganglion in the period of onset of model groups, were much more than those in control groups and in the period of intermission of model groups (p<0.05). Secondly, painful behavior of rats in Sham or E2-R was more ethology than that in OVX (p<0.05). The expression of P2X3receptor in the trigeminal ganglion in Sham or E2-R was much less than that in OVX (p<0.05). Reversely, the expression of P2Y1receptor in the trigeminal ganglion in Sham or E2-R much more than that in OVX (p<0.05)CONCLUSIONS1The low expression of P2Y1receptor and the high expression of P2X3receptor in the trigeminal ganglion might induce the attack of migraine, in model of migraine rats.2. Estrogen might mitigate the attack of migraine, by upregulating the expression of P2Y1receptor and inhitting expression of P2X3receptor in trigeminal ganglion, in model of migraine rats. |
