Objcctivc: To inveshgate the effects and mechedsm of the Chinese ComPoundPrescription Kangliu caPulse (KLC) on aPoptosis and differntiation of humanhepatocarcinoma cell line SMMC-772l. Mcthods: By using the method ofphannacology of the serum with Chinese drugs (PSCD ), the KLC drug-serum wasincubated with culfored SMMC-772l cells. The cell morphology, proliferation,agarose gel electrophoresis of DNA, the rate of aPoptosis and cell cycle phasedistribution, the expressiOn ofbcl-2 gene protein, the secretion of AFP and the Y--GT activity were observed. Results: After trCating with the drug-Senun, theproliferation of SMMC-7721 cells was inhibited evidentiy. The cells showedcharacters of aPoptosis and differentiation in cytomorphology There werecharacteristic DNA "ladder"on agarose gel electroPhoresis. Flow cytOmeny analysisshowed tyPical peaks of aPoptosis in DNA histograIn, and the rates of aPoptosis ofligh dose KLC and low dose KLC were 20.4% and l8.6% respectively, the celIpercent of S phase was increased and tha of GdGl phase and G2M phase weredecreased. Compared with the contrOl, the expression of bcl-2 gene protein. -thesecretion of AFP and the Y --GT activity were decresed obviously Conclusion:KLC could induce the apoPtosis and diffeentiation of SMMC-7721 cell, Which maybe the imPota mechedsms of KLC' anitUInor effects.
