Alzheimer's Disease (AD) is an n active field in the research of western integrated with traditional Chinese medicines for its high morbidity and incurable prognosis. An animal model and a cellular model have been set up in this research with D-galactose. Comparing with Acricept in 0.54mg/ (Kg ?d). d dosage as a positive control drug for AD patients, the effects of Danzhi-xiaoyao San (DZXYS, a traditional Chinese prescription) on AD-like disorders have been confirmed in the animal and cellular models.It has been carried out to research the mechanism of DZXYS for treating AD model of rats dealt with D-galactose.. An AD-like model of rats has been set up with sc. D-galactose ISO.Omg/ (Kg ?d) X49d. Comparing with Acricept in 0.54mg/ (Kg ?d) dosage as a positive control drug, DZXYS in 12.636g/ (Kg ?d) . d dosage has orally been administrated for the same 49 days to treat the injury in the AD-like model of rats. The general state and the weight of the rats in four groups have been observed and recorded weekly; their behavior activities in watery maze test and in step-down test had been carried out during 42-46 day; their cerebral tissues had been obtained at 49 day. The protein content, RNA content, DNA content, SOD activity, MDA content, Ach content, AchE activity in the cerebral tissues had been detested with ultraviolet assay, the contents of monoamine transmitters or their products in the cerebral tissues had been detested with waters liquid chromatography; the histological structure with HE stain and Bax or Bcl2 antigen with immunohistochemistry of the cerebral tissues have been observed with light micrography; the ultrastrucrures of the cerebral tissues have been observed with electron micrography; the content of aldose reductase-mRNA is detested with RT-PCR. The difference was analyzed between the control rats without D-galactose, the model rats dealt with D-galactose, the model rats treated with Aricept and the model rats treated with DZXYS, it is significant as P<0.05. The results have demonstrated those as follows: ?DZXYS can moderate the worse general states in the model rats, such as less activity, less reaction, less feeding, and loss weight, without any side reaction. (2) DZXYS can reserve the learning or remembering capability of model rats from their behavior activity in watery maze test and in step-down test. 〥ZXYS can increase the total DNA content from 1.093+0.267 to 1.488 + 0.341 (PO.01) in their cerebral tissues., 瓺ZXYS can activate the SOD activity from 352.670 + 37.711 to 380.728 + 14.845CPO.05) in their cerebral tissues, can reduce MDA content from 7.878 + 0.6968 to 6.636 ?.800 (.P<0.01) in their cerebral tissues. ?DZXYS can recovery the norepinephrine content from 207.171+47.261 to 244.185+37.172 (P O.05), 5-hydroxytryptamine content from 924.670 + 543.509 to 1199.77+494.871 (P O.01), 5-hydroxytryptamine ratio 0.693 + 0.118 to 0.782+ 0.060(.P<0.01), acetylcholine content from 0.677 + 0.123 to 0.969 + 0.210 (PO.05) in their cerebral tissues , and can also limit the acetylcholinesterase activity from 157.998+24.325 to 120.869+44.732 (P<0.05) in their cerebral tissues. 〥ZXYS can decrease the lesion in their cerebral tissues, such as neuron degeneration and death, glial proliferation, blood vessel wall thickening under light micrography. DZXYS can normalize the disarranged microtubule and microfilament in their neurites of model cerebral tissue; it can increase the quantity, volume, density of the mitochondria and lysosomes in their neurites of model cerebral tissue; it can omitted the autophagolysosomes and lipofuscins in their neurites in model cerebria. Under light micrography, it can also make the endothelia to proliferate and activate without damage, the basal membrane to keep fit in shape and density, and the footplates of astrocytes on the basal membranes to normalize themselves in the model cerebral capillaries. @DZXYS can overturn the decrease Bax-protein expressed amplification, and increase Bcl-2-protein expressed amplification in the model cerebral or liver tissues. 瓺ZXY...
