By means of advanced modern technology such as cDNA microarry.. cell culture flow cytometryN immunohistochemistry the sera containing drugs computer image analysis system etc, we have observed that the effeccts of WSG on inhibitories of Lewis Lung carcinoma Sarcoma-180 SPC-A₁A2 SMMC-7721 etc respectively. The experimental study was expected to explain what is the pharmacological effects and mechanism of WSG on anti-lung cancer.The results showed that: (1) WSG exhibited obviously repressive roles for several types of tumor cell straits, including Lewis Lung cavcinoma S₁₈₀ in vivo; SPC-A₁ A2 and SMCC-7721 in vitro. Meanwhile, It could be used to prolonged the survial guality of burdening tumor mice in ascites style.(2)Major anti-tumor associated targed genes using cDNA microarry technigue showed following behaviors:The genes(CD CCT) related to invasion metastasis and proliferation of malignant carcinoma were down-regulated; The genes (CytC MUP) related to apoptosis and negatively marker of malignant carcinoma were up-regulated. (3)WSG could decrease MVD of tumor tissues, and inhibite the expressions of VEGF and bFGF closely related to angiogensis. (4)WSG could decrease the expression of PCNA and enhance the rates of apoptosis for tumor cells.(5)WSG also promote immune-indexes andexcretion of IL-2 burdening tumor mice, up-regulate expression of autocrine TNF. (6)WSG could improve blood routine examination burdening tumor mice.These results indicates that:(1) WSG has definite anti-tumor effect on several types of tumor cell straits, especially for pulmonary carcinoma. ㊣ts mechanism is related to down-regulating gene expressions associated with invasion , metastasis and proliferation of tumor cells . up-regulating gene expressions asscciated with apoptosis of tumor cell and immune burdening tumor mice intervene formation of tumor angiogevsis inhibit proliferation of tumor cells and induce apoptosis of tumor cells. In addiation, WSG have advantages of enhancing immune function and protecting hemapoietic function.Target-genes treated Lewis Lung carcinoma by WSG have been high-througput screened with cDNA microarry technigus . The experimental results play some important roles in revealing WSG to anti-tumor molecular mechanism at genome level and developing scientific theories of TCM on anti-tumor.
