The root of aubergine is one of the popular meadicine of Tujia nationality. It has the function of expelling wind and removing dampness, clearing away heat and stopping bleeding, dissipating blood stasis and promoting the subsidence of swelling. It is mainly used to treat toothache, arthralgia due to wind-dampness-heat, hemafelia, beriberi, dysentery with bloody stool, chilblain etc. It has a lot of ingredients including phenols, organic acid, alkaloid, saponin, tannic etc. The reports dealing with its anti-inflammatory action are very scarce. Moreover there are not reports about its acidic components anti- inflammatory effect and the mechanism now. This thesis carried out a thoroughly and systemically research on the antiseptic effect, anti-inflammatory effect and the mechanism of the acidic components of aubergine root (ACAR) for the first time. The methods adopted include cell culture, flow cytometry, radioimmunoassay (RIA), drug sensitivity test, HPLC etc. by in vio and vitr experiment, from the level of entire animal, tissue and organ, cell and molecule. The results are as follows:1. ACAR can inhibit the acute inflammatory model characterized by increased capillary permeadillity, swelling or the chronic model characterized by hyperplasia of granulation and the chronic peridentitis of rats. It proves that the anti- inflammation effect of ACAR is clear-cut for the acute or chronic inflammation, which coincides with its function of expelling wind and removing dampness, dissipating blood stasis and promoting the subsidence of swelling. ACAR is the one of the important substantial basis of aubergine root's pharmacological effects.2. ACAR has anti-anaerobic-bacterial effect, It has the antibacterial effect toward many oral dental caries pathogenic bacteria and periodonal pathogenic bacteria. The MIC and MBC are no less then 50mg / ml for oral dental caries pathogenic bacteria, the MIC from 3.1 to 25mg / ml, the MBC from 6.2 to 50mg/ml for periodonal pathogenic bacteria.3.The anti- inflammatory mechanism of ACAR takes effect by the following means:(1)Inhibit the production and activity of COX2 in AA and decrease the production of PGE2,meanwhile reduce the content of LTB4 produced through 5-LOX. (2) Inhibit the free-radical production such as NO.MDA by Antioxidation.(3)decrease the content of inflammatory cytokines as TNF,IL-6,IL-8.(4) Inhibit the activation of NF- B.4.The realization of anti- inflammation of ACAR is not by enhancing the function of hypothalamus-pituitary-adreno, but is some kind of direct influence.The results indicated that ACAR inhibit the activation of NF- B during inflammation, and further prevent the gene expression and protein secretion of cytokine, some enzymes associated with inflammation. It can inhibit the secrtion and discharge of inflammation media, thus interdict the destruction of periodontal tissue and the absorption of alveolar. As a result, it controls the inflammation and treats the peridentitis effectively.
