[Background] Left ventricular hypertrophy (LVH) is one of the most common complications of hypertension, which is not only the results of compensated cardiac tissue growth due to heart overload, but also the pathological process of heart remodeling. Epidemiological evidence showed that hypertension with LVH relates to the significant increase of morbidity and mortality associated with cardiac sudden death, arrhythmias, angina pectoris, myocardial infarction, heart failure, and other cardiovascular diseases, which affects the prognosis of patient. LVH is an independent risk factor of cardiovascular diseases now. It is important to discover mechanisms of LVH and to find preventive measures, which have significance in the field of both theoretic studies and clinical practice. More recently, rennin-angiotensin system (RAS) activation has been understood to be a major inducer of cardiac tissue hypertrophy. It has been demonstrated that the local cardiac angiotensin II is a potent growth factor in LVH, which can be blocked by AT1 receptor antagonists.There is no LVH of hypertension in Chinese medicine, but the basic knowledges of a series of changes of LVH of hypertension and body of human health are balance and imbalance of yin and yan in human body based on basic theory of Chinese medicine. The body of human health is based on the yin and yan interdependence for each other, such as yin wanes, yang waxes, and vice versa. A series of complications of hypertension result from imbalance of yin and yan, while LVH of hypertension is formed new relative balance under new circumstances. The relative balance is the compensated results of preceding imbalance of yin and yan. The pathologic basis of LVH is blood stasis, qi deficiency and yin insufficiency of liver and kidney, especially the blood stasis. The treatments of drugs are promoting blood circulation, reinforcing qi, replenishing kidney, tonifying liver and nourishing yin. Such treatments get some clinic improvement of patients.[Objectives] The objectives are to discovery some Qi tonic and drugs for promoting blood circulation of Chinese herbs who can prevent the cardiac hypertrophy and study their mechanisms of cardiac hypertrophy induced by Ang by researching the effects of Qi tonic and drugs for promoting blood circulation of Chinese herbs on cardiac hypertrophy and related genes induced by Ang.[Methods] The models of the myocardial hypertrophy and proliferation of the cardiac fibroblasts induced by angiotensin were established by observing the content of the total cellular protein, cell diameter, and the number of the cells in the cultured neonatal myocardial cells in vitro.In addition to the above parameters, the studies were also done to understand the effects of Qi tonic and drugs for promoting blood circulation of Chinese herbs on the myocardial hypertrophy and proliferation of the cardiac fibroblasts induced by angiotensin II in the cultured neonatal myocardial cells in vitro. The efficient components of Danshensu, Ligustrazine, and Astrgalus Root injection were elected, while serum pharmacology of Dangshen and Dangshen plus Danshensu were used.In order not to damage the cultured neonatal myocardial cells in vitro, there were three different serum concentrations of Dangshen and Dangshen plus Danshensu in the experiment of MTT.Research groups: control group, Ang group, Ang plus Losartan group, Ang II plus three different serum concentrations of Danshensu (large, middle, small) group, Ang plus three different serum concentrations of Danshensu (large, middle, small) group, Ang pius three different serum concentrations of Ligustrazine (large, middle, small) group, Ang plus three different serum concentrations of Astrgalus Root (large, middle, small) group, Ang plus three different serum concentrations of Dangshen (large, middle, small) group, Ang II plus three different serum concentrations of Dangshen plus Danshensu (large, middle, small) group.Ca2+ in myocardial cells and myocardial fibroblast was detected by coolter EPICS XL with Fura-3, in order t...
