Object: Through the study of huatanfang which can affect Epidermal growth factor receptor (EGFR) to synthesis of mucins, we hope reveal the molecular essence of huatanfang which elimated sputum.Methods:(l)forty-eight rats were divided into three groups randomly: normal group, model group and treatment group. We established rat's model of chronic bronchitis by intratracheal instillation of lipopolysaccharide 0.2mg. Test of phenolsulfonphthalein elimating sputum was tested. (2) The rat's lung tissue section stained by HE was observed to study the morphological alternations. (3) Using reverse transcription polymerase chain reaction (RT-PCR),the expression of MUC5ACmRNA in lung tissues was tested. The expression of MUC5AC was tested with the method of immunohistochemistry. (4) EGFR was tested with with the method of immunohistochemistry. (5) After the mice were killed, theirleft lungs were taken out for bronchi-alveoli fluid, then the percentage of PMN was worked out. (6)Tumornecrosis factor- α (TNF-α) in blood serum of rats were measured with enzyme-linked immunoadsordent assay(ELISA).(7) The expression of nuclearfactor-kappaB(NF-κB) in lung tissues was tested with the method of immunohistochemistry.Results:(1) The volume of elimating sputum of the model group is higher than that of the normal group (P<0.01). The level of the treatment group obviously decreased, which was very markedly different from that of the model group (P<0.05). (2) There are the features of goblet cells, submucosal glands hyperplasia and hypertrophy in model group. The above-mentioned lung damage could be rehabilitated through treating of Chinnese pharmacy. (3) The expression of MUC5AC mRNA and MUC5AC is higher than that of the normal group (P<0.05). The level of the treatment group obviously decreased, which was very different from that of the model group (P<0.05). (4) The expression of EGFR in lung tissues is higher than that of the normal group (P<0.01). The level of the treatment group obviously decreased, which was very markedly different from that of the model group (P<0.01). (5) Thenumber of PMN of the BALF in the model group is higher than that of the normal group (P<0.01). The level of the treatment group obviously decreased, which was very markedly different from that of the model group (P<0.01). (6) The level of blood serums TNF- a of the model group is higher than that of the normal group (P<0.01). The level of the treatment group obviously decreased, which was very markedly different from that of the model group (P<0.05). (7)The expression of NF- k B of the model group in lung tissues is higher than that of the normal group (P<0.01). The level of the treatment group obviously decreased, which was very markedly different from that of the model group (P<0.01).Conclusion: (1) Molecular essence of huatanfang which elimated sputum decreased the expression of EGFR and mucins. (2) Huatanfang can decrease the expression of mucins by decreasing the number of PMN and TNF- α. (3) Huatanfang can decrease the expression of mucins by decreasing the expression of NF-κB.
