Glucose deprivation plays an essential role in the pathogenesis of tissue damage, and cell dysfunction caused by ischemia. The investigation of molecular and cellular changes in the process of glucose deprivation will contribute to understand the pathogenesis of these diseases. And the researching of the effective molecular interference to inhibit the cell damage induced by glucose deprivation is useful. grp75, a member of the hsp70 family of proteins is a kind of molecular chaperon of mitochondria.The researching work of our lab demonstrated that the overexpression of grp75 showed a significant protective effect against cell glucose deprivation.The expression of grp75 recently has become a major interest of clinicians because it has been detected in a clinical settings of ischemia. Further, searching one kind of security drugs which can upregulate the expression of grp75 would be a clinical implications. Recent, Paul E. Wischmeyer and his colleagues indicate that a single dose of intravenous Gln can enhance hsp expression,decrease end-organ injury, and enhance survival from septic shock in the intact rat. Thus,we observed the effect of glutamine to grp75(the member of the hsp family).Data indicated that Gln can upreglated the expression of grp75 in vitro model.But the effect seems not related with the dose and effective time of Gln. And the result of RT-PCR and in situ hybridization showed that mRNA level of grp75 is increased under the effect of Gln, these results suggested the possible mechanism by Gln to enhances the expression of grp75 is to administrate the transcription activity of grp75.In order to observe the protective effect of Gin to cell damage induced byglucose deprivation and to animal brain ischemia, and to research the relationship between the protective effect of Gln and grp75,we built in vitro and in vive model to imitate ischemia. MTT, and flow cytometry analysis was taked to detect the growth rate and apoptosis rate of PC 12 cell. It showed the growth rate of the cells cultured with no-glucose media is as 26% as normal cells, and the apopotosis rate is 33.42%.The cell growth rate is markedly increased and the apoptosis rate is decreased after Gln was added to the media.The effect of Gln to increased growth rate of cells under glucose deprivation is decreased when anti-grp75 was transfected to the cells,it proved that Gln is a enhancer to upreglate expression of grp75, and it appears to be...
