The changes in tubular epithelial cells play an important role in the progression of renal insufficiency. The compensatory hypertrophy of thenephron , especially the proximal convoluted tubule , is a classic theory to explain the mechanism of the progression of chronic renal insufficiency. The preliminary studies in recent years suggested that tubular epithelial cells may undergo the transdifferentiation to myofibroblast during the process of the progression of interstitial fibrosis to directly contribute the accumulation of extracellular matrix in the interstitium. The exact effect of A II on the tubular epithelial cell has not been fully elucidated , especially the effect of A II on the transdifferentiation of tubular epithelial cells to myofibroblast. We investigated the role of A II and A II type 1 receptor antagonist (ATlRa) valsartan on the growth and transdifferentiation of tubular epithelial cells in porcine tubular epithelial cell line LLC-PK1 in this study .METHOD AND RESULT : The DAN , RNA and protein synthesis of LLC-PK1 cells were measured by 3H-thymidine , 3H-uridine and 3H-proline incorporation to access the proliferation and hypertrophy of these cells incubated with A II and/or ATlRa. The transdifferentiation of LLC-PK1 cells was measured by immunohistochemistry using anri-vimentin and anti-a-smooth muscle actin(α-SMA) monoclonal antibodies . The intracellular calcium mobilization was measured dynamically by laser confocus microscopy using Fluo-3 molecular probe and the expression of TGF-β1 mRNA was measured by RT-PCR.Measured by 3H-thymidine incorporation in LLC-PK1 cells at 24 , 48 and 72 hours , there was no effect of A II (10-10. 10-8 and 10-6M) on the proliferation of LLC-PK1 cells.
