Pancreatic islet transplantation has been the ideal therapeutic approach for type 1 diabetes, whereas its long-term effect is not very optimistic,it is suspected that the failure of islet transplantation has been closely related to cells composition of grafts. Our study investigated the effects of pancreatic a cell, a cell conditional media and GLP-1 on insulin releasing and biosynthesis function by co-incubation of pancreatic immortal a cells line and β cells line, aiming to find out the proper proportion of a cells and β cells in grafts at which pancreatic a cells regulate β cells most effectively, therefore achieve the best transplantation effects so that the transplanted pantients can acquire the physiological insulin releasing as the healthy, thus control the disturbance of diabetes for a long time and improve the life quality of diabetic patients.ObjectiveTo investigate the effects of pancreatic a cell , a cell conditional media and exogenous rhGLP-1(7-36) on insulin releasing and insulin messenger RNA expression level in pancreatic β cell (INS-1).Methods1. The acute insulin secretion experiment were practiced in INS-1 cells by co-incubation ①with different proportion of pancreatic a cell (the number of a cells is 0%,3%,10%,30% ) for different incubation time(30,60,120,240 minutes) at the presence of 20mmol/L glucose concentration. ② with the different proportion of pancreatic a cell (different proportion. 0%,3%,10%,30%) for 240 minutes at the presence of different glucose concentration(final concentration is 1.85,5.6,16.7,50mmol/L,respectively).
