Objective To analyze the expression of KAI/CD82 and its relationship with clinicopathologic characteristics of hypopharyngeal carcinoma, such as tumor stages, cell differentiation and lymph node metastasis status. To explore the biological function of KAI1 gene in lymphatic metastasis of hypopharyngeal carcinoma and its relationship with mutant p53 protein. To evaluate its correlation with prognosis of hypopharyngeal carcinoma.Methods 52 cases of paraffin-embeded hypopharyngeal squamous cell carcinoma were collected. Immunohistochemistry SABC method was performed to examine the expression level of KAI1/CD82 and p53 protein, the relations between their expression and clinicopathologic characteristics (metastasis status, tumor stage, grade of tumor differentiation and prognosis) were evaluated by x~2 analyses. Odds ratios(OR) and 95% confidence interval (95%CI) were calculated by binary logistic regression to evaluate the risk factor of tumor metastasis. 40 cases of fresh hypopharyngeal cancer samples and 12 cases of normal pharyngeal tissues were examined by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), the correlations between KAI1mRNA expression and pathologic characteristics were also analysed.Results1.
