| Thalictrum atriplex finet et Gagnep.(Fam.Ranunculaceae)is a Chinese folk and Tibetan medicine, found mainly on the plateau of 23003600 meters above sea level in southwestern China. The roots of the plant called "Ma-Wei-Lian", or "Shui-Huang-Lian" are used for the treatment of infectious hepatitis, carbuncles, dysenteric diarrhea, and certain gastroenteric disorders. The genus Thalictrum consists of about 200 species worldwide, among which 67 are distributed throughout China, but mostly in the southwestern region.In this thesis, a systematic phytochemical study on the constituents of Thalictrum atriplex including both alkaloid and non-alkaloid has been carried out for the first time. Several of the alkaloids isolated have been screen tested on three pharmacological activities. In addition, the distribution pattern and chemotaxonomic significance on aporphine-benzylisoquinoline(ABI) alkaloids within the genus have been discussed.Thirty-one compounds were isolated and identified from the ethanol extraction of aerial part (stems and leaves) of the plant for the first time. Twenty-two isoquinoline alkaloids, of which six aporphine, ten aporphine-benzylisoquinoline(ABI), three bisbenzylisoquinoline(BBI) and three other type of isoquinoline alkaloids, were obtained from ether and chloroform extraction. Among them there are three new ABI alkaloids ,named thaliatriplexine ( VIII,XIV ) ,thaliatriplextine ( XVII ) and thaliatriplexonine(XXVII). Thaliatriplexonine , the moiety of its aporphine is in the quaternary state on structure, is the first quaternary ABI dimmers. The non-alkaloid fraction, i.e. the ethyl acetate extraction ,was also studied and lead to the isolation and structure determination of nine other compounds. Three of them are flavonoids. One flavonol glycoside named kaempferol-3-O-[3'''-acetyl-arabinopyranosyl(1'''-6")]-β-D-glucopyranoside (No.38) was confirmed as a new constituent.Seven compounds were isolated from the chloroform fraction of ethanol extraction from the underground part (roots) of the plant. Three new bisbenzylisoquinoline alkaloids named northalfine (R-I). northalfinine (R-III) and thaliatrine (R-IV), together with three known dimeric alkaloids are all obtained from the roots for the first time.The spectral features and characters of thalifaberine tape of Aporphine-benzylisoquinoline (ABI) alkaloids on UV,IR, MS, 1HNMR and 13CNMR were summarized up according to those spectral data of compounds we isolated and published spectral data.On the basis of above phytochemical research, fifteen different alkaloids... |
