Objective: To explpore the effect of XuanFu DaiZhe decoction on the contraction of the rat's gastric smooth muscle cell (GSMC) and signal transduction pathway.Methods: The serum containming XuanFu DaiZhe decoction and the serum without XuanFu DaiZhe decoction were taken from the rats. Single gastric smooth muscle cells were isolated with collagenase from three parts (gastric antrum, gastric body and pylorus)of the rat's stomach. Based on the method, the effect of XuanFu DaiZhe decoction on the contraction of the rat's GSMCs was observed and the effect of XuanFu DaiZhe decoction on the contraction of the rat's GSMCs after adding U-73122 (a phospholipase C inhibitor), heparin (IP3 receptor antagonist) and TMB-8 (intracellular Ca2+release inhibitor) respectively was observed. The rat's GSMCs were cultured in vitro by collagenase digestive method. The method of immunohistochemistry assay was uesd to detect the expression of protein kinase C (PKC). The method of in situ hybridization was used to detect the expression of cholecystokinin B/gastrin receptor mRNA(CCKB/GASRmRNA), vasoactive intestinal peptide receptor2 mRNA(VIPR2mRNA) and inositol-1,4,5- triphosphate receptor mRNA(IP3R mRNA).Results:1. The serum containming XuanFu DaiZhe decoction could cause the rat's GSMC to contract.2. The serum containming XuanFu DaiZhe decoction induced contraction of gastric antral smooth muscle cells were inhibited by U-73122, heparin and TMB-8 in some extent.3. The serum containming XuanFu DaiZhe decoction groups could increase the expression of CCKB/GASRmRNA of the rat's GSMCs and reduce the expression of VIPR2mRNA.4. The serum containming XuanFu DaiZhe decoction groups could increase the protein expression of PKC of the rat's GSMCs.5. The serum containming XuanFu DaiZhe decoction groups could increase the expression of IP3RmRNA of the rat's GSMCs.Conclusion: (1)XuanFu DaiZhe decoction can induce obvious contraction of the rat's GSMCs. (2)Through activating the CCKB/GASR, XuanFu DaiZhe decoction can increase the secretion of CCK/GAS and Through repressing the VIPR2, XuanFu DaiZhe decoction can decrease the secretion of VIP, which can promote the contraction of the GSMCs. (3)Through activating the PKC in the GSMCs, XuanFu DaiZhe decoction can regulate calciumion(Ca2+) to promote the contraction of the GSMCs. (4)Through activating IP3R, XuanFu DaiZhe decoction can regulate calciumion(Ca2+) to promote the contraction of the GSMCs.
