Objective: To discuss the expression and role of matrix metalloproteinases and tissue type inhibitors of metallproteinases and transforming growth factor beta during the aging in the kidney of rats. Meantime, to investigate the effect on prolong kidney aging by moxibustion the pointsShenshu(BL23).Methods: On the base of nature aging rats model, the rats were divided into 3 groups: adults group, aging group, and moxibusition aging group, the latter were treated by moxibustion Shenshu(BL23) points in 10min, one a day, 5days a course of treatment, all 8 courses. Using immunohistochemical method to observe each group's alteration on the kidney tissue, and to test some signs' change on MMP-2, MMP-9, TIMP-1, TIMP-2, TGF-βprotein.Result: the expression of TIMP-1, TIMP-2 and TGF-β1 was mainly in the glomeruli, and tubulointerstitial, and the levels of theirs expressions were increased with aging(P<0.01). MMP-2 and MMP-9 was detected in tubular epithelial cells and no change of their expression was found during aging. Through moxibustion 8courses on Baihui(DU20), Shenshu(BL23), compare to aging group, the moxi-aging group has distinctive difference(P<0.01) in all signs.Conclusion: The cell factor TGF-β1 increasing promotes the expression of TIMP-1, TIMP-2 in aging kidney, which inhibit the hydrolysis activity of MMP-9, MMP-2 protein, and due to Extracelluar Matrix's compound and hydrolysis improving from balance to unbalance, so it lead glomerulosclerosis and interstitial fibrosis to occur. Above all, which shows the imbalance between the expression of matrix metalloproteinases and tissue type inhibitors of metalloproteinases may play an important role in the process of kidney aging. While moxibustion on Shenshu may not only significantly decrease the expression of TGF-β1, TIMP-1, TIMP-2, and correspoundingly regulate the ratio of MMP-9/TIMP-1, MMP-2/TIMP-2, but also effectively prolong glomerulosclerosis and interstitial fibrosis, so as to improve and protect kidney's structure and function.
