| Object:Our research focused on expression of CD80/CD86 of decidua tissue in abortion rats model with renal deficiency and Th2-type immunity deviation in abortion rats induced by use of ZhuyunⅢto improve the result of pregnancy,and some proofs, which proved Chinese medicine theory of "kidney controls reproduction",were founded as well.Methods:SPF female rats,10 weeks old,weight 200~250g were distributed into 6 groups at random,which were group A:control,B:model,C:model+medicine, D:model+low dosage TCM,E:model+middle dosage TCM,F:model+high dosage TCM,to observe vaginal smear.abortion rats model with renal deficiency was made by intragastric administration of hydroxycarbamide 300mg/kg/D in dioestrus. Mifepristone(RU486)was used by intragastric administration in 8thday,rats were killed after 24 hours,and the following data was measured:embryos number,abortion rate and diameter of embryos.Decidua tissue on one side was collected in 1 hour after rats death in asepsis condition to detect expression of CD80,CD86,CD28,CTLA-4 with flow cytometry. Decidua tissue on the other side was embedded and frozen.MRNA Expression including Th1(TNF-α,IFN-γ,IL-2)and Th2(IL-4,IL-6,IL-10)was detected by semiquantitative RT-PCR.Results:1.Intake of food/water:(1)compared with control group,intake of food in model group was reduced markedly in 4thand 8thday(P<0.05)(2)compared with model group,changes of intake of food in high dosage group were significant in 8th day(P<0.05)(3)compared with abortion model group,changes of intake of water were significant in control,medicine,middle dosage herb and high dosage herb groups(P<0.05).2.Changes of pregnant rats body weight in 4thand 8thday(△W2-W1):There was no significant△W2-W1change in all groups in 4thday(P>0.05).Body weight improvement indicated in 8thday:control group>high dosage group>medicine group>middle dosage group>low dosage group>model group,and the difference of body weight improvement in model group was significant compared with other groups in 8thday(P<0.05).Compared with model group,body weight improvement in middle dosage group,high dosage group and medicine group were significant (P<0.05)in 8thday.3.Organ index in pregnant rats:uterus organ index in model group was the lowest,the difference was significant compared with A,C,E,F groups (P<0.05).There was no significant difference in kidney and ovary index in all groups(P>0.05).4.Embryo diameter and abortion rate of pregnant rats:There was no significant difference on embryo diameter between model group and other groups(P>0.05). There was significant difference on abortion rate between model group and other groups(P<0.05).Number of alive embryo in B,C,D and E groups was significantly different from A group(P<0.01).Abortion rate in high dosage group,middle dosage group and medicine group was significantly different from model group(P<0.05).5.Expression of costimulatory molecule of fetomaternal interface:Expression of CD80,CD86,CD28 in normal rats embryo was low,and that of CTLA-4 was high. expression of CD80,CD86,CD28 in abortion rats model with renal deficiency inclined,and that of CTLA-4 declined.In kidney deficiency abortion model groups, expression of CD80,CD86,CD28 declined after therapy in medicine and high dosage groups,the difference was significant(P<0.05),however,expression of CTLA-4 improved significantly(P<0.05).6.In abortion rats model with renal deficiency,expression of Th1(TNF-α,IFN-γ,IL-2)improved and that of Th2(IL-4,IL-10)declined.ZhuyunⅢcould inhibit expression of Th1 and promote expression of Th2 in model group.Conclusion:1.It may be the reason of abortion and upgrading of abortion rate that included high expression of CD80,CD86 and CD28,low expression of CTLA-4,which inhibit as well as high expression of Th1 and low expression of Th2.2.It may be mechanism of anti-abortion with ZhunyunⅢthat reduced expression of CD80/CD86 and CD28 and improved expression of CTLA-4,which inhibited costimulatory signals that were produced by combination of CD28 and CD80/CD86,then inhibited expression of Th1 and promoted expression of Th2 so that could regulate unbalance of cytokines,which was caused by high expression of Th1 and low expression of Th2,in embryo of abortion rats.Moreover,ZhunyunⅢcould reduce immunologic rejection and protect embryo,decline abortion rate in abortion rats model with renal deficiency and treat abortion,its effect was as good as progestogen.Our research proved that ZhuyunⅢcould treat abortion. |
