| Objective:To establish the rats model of severe acute pancreatitis(SAP)induced by sodium cholate sulfur and confirm its success so as to study on changes of Ghrelin and GHSR density in serum and pancreatic tissue and the influence of Dan Shen in the SAP rats model to investigate the it's mechanism of treatment on serve acute pancreatitis.Methods:The study was performed on 48 Wistar rats.Two experimental groups composed 24 rats were established:acute severe pancreatitis(SAP)group, sodium cholate sulfur was injected intrapancreatobilliary duct by using I.V.Catheter and microsyringe.The operated rats were injected NS in the post period.Control group,deionized water was injected intrapancreatobilliary duct by using I.V.Catheter and microsyringe.Six animals in each subset were sacrificed after taking the blood sample,the pancreas,at the time of 3h,6h,12h,24h after cessation of Sodium taurocholate injections.To detect the level of serum amylase,TNF-αand IL-6,take pancreas tissue to evaluate the existence of pathological changes of pancreas to evaluate the existence of severe acute pancreatitis.Seventy-two Wister rats were randomly divided into 3 groups i e,sham operation group,SAP model group,danshen treatment group,twenty-four rats were in each group.Each group was subdivided into 4 subsets,six rats were in each subsets,called three hours,six hours,twelve hours, twenty-four hours subset.SAP model group,sodium cholate sulfur was injected intrapancreatobilliary duct by using I.V.catheter and microsyringe,sham operation group,deionized water was injected intrapancreatobilliary duct by using I.V.Catheter and microsyringe.DanShen treatment group,after the establishment of SAP model,was treated with Danshen peritoneal injection.The other two group were treated with NS peritoneal injection.After the opration,at the corresponding time, each subset was taken the blood from the heart,then,was killed and pancreas was taken from the body.So,we can detect the level of Ghrelin in serum,and using RT-PCR to detect the expression of Ghrelin mRNA and GHSR mRNA,using western blot to detect the expression of Ghrelin and GHSR protein.Result:①Sodium taurocholate led to the development of severe acute pancreatitis.The model rats pancreas Structure was destroyed.The pathological scores of the SAP groups were significantly higher than the control groups(P＜0.05).The levels of TNF-αand IL-6 in SAP groups markedly decreased than the controls(P＜0.05),and then decreased to a level closed to that of the control group at 24h.The levels of serum amylase in SAP groups were obvious higher than the controls(P＜0.05).②the Ghrelin density of serum is deferent among sham operation group,SAP model group and danshen treatment group.The deference is statistical(p＜0.05).the Ghrelin density of danshen treatment group is obviously higher than that of sham operation group and SAP model group,the Ghrelin density of danshen treatment group decrease accompanying time going,in 24h subset there is no obviously deference between SAP model group and danshen treatment group.Ghrelin mRNA was expressed in pancreatic tissue,and the expression quantity of SAP model group,danshen treatment group were more than that of sham operation group,the difference was statistical(p＜0.05);the expression quantity of danshen treatment group was more than that of SAP model group;the expression quantity of the subsets in sham operation group were not obviously deferent,the Ghrelin mRNA expression quantity of the subsets in SAP model group and danshen treatment group were increasing gradually.The deference between 3h subset and 12h subset or 24h subset is obvious(p＜0.05).Ghrelin protein is expressed in pancreatic tissue,the expression quantity of SAP model group, danshen treatment group were more than that of sham operation group,the difference was statistical(p＜0.05;the expression quantity of danshen treatment group was more than that of SAP model group(P＜0.05);the expression quantity of the subsets in sham operation group were not obviously deferent,the Ghrelin protein expression quantity of the subsets in SAP model group and danshen treatment group were increasing gradually.The deference between 3h subset and 6h subset is no obvious.③GHSR mRNA was expressed in pancreatic tissue,and the expression quantity of SAP model group and danshen treatment group were more than that of sham operation group,the difference was statistical(p＜0.05);the expression quantity of danshen treatment group was more than that of SAP model group(P＜0.00);the expression quantity of the subsets in sham operation group were not obviously deferent,the GHSR mRNA expression quantity of the subsets in SAP model group and danshen treatment group were increasing gradually.The deference between 3h,6h subsets and 12h,24 subsets is obvious(p＜0.05);GHSR protein is expressed in pancreatic tissue,the expression quantity of SAP model group,danshen treatment group were more than that of sham operation group,the difference was statistical(p＜0.05);the expression quantity of danshen treatment group was more than that of SAP model group(P＜0.05);the expression quantity of the subsets in sham operation group were not obviously deferent,the GHSR protein expression quantity of the subsets in SAP model group and danshen treatment group were increasing gradually.The deference between 3h,6h subsets and 12h,24h subsets is obvious.Conclusion:①The rat SAP model were established successfuly by intracholedochal injection with sodium cholate sulfur.②Under the action of danshen the Ghrelin protein quantity in serum and pancreatic tissue increased markedly,the Ghrelin mRNA expression quantity increased gradually. Danshen can treated pancreatitis through increasing the Ghrelin expression quantity.③Under the action of danshen the GHSR protein quantity in pancreatic tissue increased markedly,the GHSR mRNA expression quantity increased gradually. Danshen can treated pancreatitis through increasing the expression quantity and the integration of GHSR with Ghrelin.
