| Objective:To investigate the effect of the Shenshuaixiezhuo Pill on FN,Col-Ⅳ,TGF-β1 of serum,urine and kidney tissue of renal interstitial fribrosis rats caused by adenine,and to illustrate the mechanism of Shenshuaixiezhuo Pill on delaying renal interstitial fibrosis.Methods:In the experiment,the rats model with renal interstitial fibrosis caused by adenine was adopted.The rats were observed for the change of the common physical order and quantity.The serum,urinary FN, Col-Ⅳ,TGF-β1 in rats were measured by two-layer antibody sandwich enzyme-linked immunosorbent assay(ELISA)method.HE and Masson staining method were used to observe the pathologic change of the experimental rats kidney tissue.The protein expression of FN,Col-Ⅳ, TGF-βand their mRNA in kidney tissue were detected respectively by immunohistochemistry with two steps,Western blot and RT-PCR.Results:The physical status and symptom of the RIF rats were improved obviously after treating by Shenshuaixiezhuo Pill;The urinary FN,Col-Ⅳ,TGF-β1,serum Col-Ⅳwere reduced and the serum FN, TGF-β1 were increased.The protein expression of FN,Col-Ⅳ,TGF-βand their mRNA in kidney tissue were reduced;Compared to comparison group,the crutive effect of Shenshuaixiezhuo Pill was significant difference.Conclusions:Shenshuaixiezhuo Pill could decrease the area of renal interstitial fribrosis through ruducing the inflammation in renal tubular; The serum,urinary FN,Col-Ⅳ,TGF-β1 were changed by reducing they production in kidney;The ECM sedimentation was reduced through depressing the protein expression of FN,Col-Ⅳ,TGF-βand their mRNA.So the renalinterstitial fibrosis was relieved by Shenshuaixiezhuo Pill. |
