| Xuan-shen is the root of Scrophularia ningpoensis Hemsl.,family Scrophulariaceae.It is a well known traditional Chinese medicine,which has the effects of cooling the blood and nourishing "yin".Xuan-shen is a China's endemic plants,mainly distributes in Zhejiang province and is one of "Zhejiang Bawei" medicine.As a result of the transfer of industry, cultivated area is constantly shrinking in Zhejiang Province.Zhenping is a mountain county in southern Shaanxi Province,adjacenting to Chongqing,is a typical sub-tropical climate.Local people had planted xuan-shen for many years,the annual output has reached to 4,000,000 tons and had exported to Southeast Asian countries and Japan,South Korea for its good quality. Zhenping became the first planting xuan-shen County,and formed a brand of "Zhenping Xuan-shen"In order to compare inherent differences in quality between xuan-shen from Zhejiang and xuan-shen from Zhenping,we studied the chemical composition and biological activity of xuan-shen from Zhenping,and we also studied the leaves of xuan-shen for a preliminary development and utilization.1 Study of Chemical composition of Xuan-shen form ZhenpingThe roots of xuan-shen were extracted with 70%alcohol.The extracts were concentrated in vacuo and then extracted in petroleum benzine,EtOAc,n-butanol,successively to give different fractions.These fractions were separated and repurified by Silica gel column and preparatived liquid chromatography to obtain eleven compounds.The structures of compounds were identified by spectral evidence of UV,IR,MS,13C NMR,1H NMR,HMBC,HMQC,H-HCOSY.These compounds were identified as iridoid glycosides, phenylpropanoid glycosides and saponins,etc,two of them were obtained from the roots of Xuan-shen for the first time.2 The quantitative analysis of iridoid glycosides in xuan-shenA HPLC-MS method was established for simultaneous quantitative determination and identification of five iridoid glycosides in different organs,different area and different months of xuan-shen;A capillary electrophoresis method(HPCE) has been developed for the simultaneous determination of four iridoid glycosides in xuan-shen.The effects on separation of buffer pH,buffer concentration,and organic modifiers were investigated.The extracts of S. ningpoensis were well separated within 45 min;Samples were prepared under different tempreture(40℃-120℃) from the fresh roots,a HPLC method was used to study the drying temperature'effect on the content of the iridoid glycosides,it was found that 100℃can mostly save the iridoid glycosides in xuan-shen.3 Studied the biological activities and application of Xuan-shen from ZhengpingThe antimicrobial activity of the extracts of xuan-shen and substances of xuan-shen were studied in vitro against eight reference strains of bacteria by using disc-diffusion method and Micro-well dilution assay.The result shown the leaves extract and Scrokoelziside A had significant activity for beta-hemolytic streptococci(CMCC,32210),The MIC and MBC were 7.81 mg/mL and 31.25 mg/mL for the leaves extract and 1.55 mg·mL-1 and 6.25 mg·mL-1 for Scrokoelziside A:The antioxidant activity was studed through scavenging effects to DPPH radical,two flavonoid glycosides in leaves have significant antioxidant activity. Scavenging effect of Nepitrin was high as well as Vitamin C;The pharmacokinetics of 6-O-methylcatalpol was studied and the plasma concentration profiles of it were best fitted by the two-compartment model after intravenous(ⅳ) administration in rabbits.In addition,the partition coefficient of the iridoid glycosides between n-octanol and water was determined according to the shake-flask method.The inherent differences in quality between Xuan-shen from Zhejiang and Xuan-shen from Zhenping were compared by studying the chemical composition and biological activity. The result of this experiment shown the types of chemical composition had no difference between each other,but the total content of four iridoid glycosides in xuanshen from Zhenping were higher than Xuanshen from Zhejiang,the further anti-inflammatory and analgesic experiments shown xuanshen from Zhenping we studied was slightly better too. This is maybe due to different climate,environment and processing.Our experient shown the good quality of xuanshen from Zhenping,and the resault provided a scientific basis for large-scale cultivation in Zhenping. |
