Background:Breast cancer is one of the major malignant diseases in women, which seriously threats to women's health.Early screening is of great significance for diagnosis,treatment and prognosis in breast cancer.This study tries to establish the serum proteins screening model for breast cancer.Method:The serum samples of 43 patients newly diagnosed with breast cancer and 45 controls were analyzed by surface-enhanced laser desorption/ ionization time-of-flight mass spectrometry.Protein peaks were collected from samples of 30 breast cancer patients and 30 controls and serum screening model for breast cancer was established by software BPS5.0.The model was evaluated by masked test of the last 13 breast cancer samples and 15 controls. All samples were analyzed according to pathological stage and lymph node classification.Results:A total 28 protein peaks was significantly different between breast cancer and control.Two peaks M/Z:9292.632 and M/Z:15131.99 were consistent with two proteins which have identified by serum protein finger print profile. M/Z:5635.228 and M/Z:3935.801 were chosen to establish screening model. The model was composed of 2 nodes and 3 terminal nodes.Its sensitivity and specificity were 92.85%and 93.33%.In the 41 clear staged breast cancer patients,protein peak M/Z:5635.33 had greatest difference in all stages. In the 41 clear lymph-node-classification breast cancer patients,protein peak M/Z:3935.959 had greatest difference in three groups.Conclusion:Serum protein mass spectrometry is a platform of high precision detective equipment combination of intelligent software.This technique makes sense for breast cancer diagnosis and research for its natural processes.
