Asthma is a mutifactorial disease,with both genetic and environmental factors contributing to its pathogenesis.Since the first genome-wide screen for asthma and atopy susceptibility loci,hundreds of asthma-related genes were found.Until now,six asthma genes were identified through the approach of positional cloning.Both of DPP10 and PHF11 were identified as asthma genes in 2003 by William Cookson's research group. Our research is the first association study between DPP10,PHF11 gene polymorphisms and asthma susceptibility in Chinese patients with asthma.This case-control study has successfully recruited 407 asthmatic patients and 288 health controls.The diagnosis of asthma was given by the experienced respiratory expert according to the ATS criteria. Pulmonary functional test results,serum IgE and peripheral eosinophil counts were collected in the asthmatic group.11 SNPs in DPP10 and PHF11 were tested and the genotyping data were analyzed by statistical tools.The results were:PHF11 B52,B53, 4459315bp polymorphisms were associated with asthma,the P values were 0.056,0.035, 0.052 respectively.PHF11 B11 polymorphism was significantly associated with eosinophil level(%),P value was 0.0078.The PHF11 haplotypes AATA15bpA, AACA15bpA were associated with asthma,P values were 0.0419,0.0124 respectively. DPP10 91P polymorphism was significantly associated with LnIgE and eosinophil level (%),P values were 0.00032,0.0023 respectively.Our study provides further support for the important roles of PHF11 and DPP10 in asthma susceptibility.
