| Objective: To observe the interference of Yu-kui Ointment, a TCM drug which can regulate qi and promote blood flow and remove stagnation and promote granulation, in assisting formation of blood vessels of rats with skin ulcer and in cells proliferation / apoptosis, and to study the healing mechanism of surface ulcers,. Method: 150 SPF Wistar rats were used in the experiment to make a rat pattern by adopting plastic ring granuloma quantifying, and were divided randomly into five groups: the simply diseased model group, the group treated by Granulation-promoting and Skin-healing Ointment, three groups treated by Yu-kui Ointment in a small, medium, large dose respectively. After they were treated by the respective medicine the weight of granuloma in each group was examined and in immunohistochemical way we detected the expression and apoptosis of VEGF,bFGF,PCNA,Bcl-2,bax in granuloma tissue. Result: The weight of granuloma increased in the three groups treated by Yukuigao Ointment. The immunohistochemical result shows that Yukuigao Ointment can increase the expression of VEGF,bFGF, and accelerate the formation of new blood vessels. The two groups treated by Yu-kuigao Ointment in medium and large doses had better results than the group treated by Skin-healing Ointment. Besides, Yukuigao Ointment can promote the expression of PCNA and promote the proliferation of wound tissue cells. Furthermore, Yukuigao Ointment can influence the expression of apoptosis-related factors—bcl-2/bax, thereby reducing apoptosis. Conclusion: Yukuigao Ointment can regulate qi, promote blood flow, remove stagnation and promote granulation. It is effective in healing of skin ulcer, which is gained by increasing the expression of VEGF, bFGF and PCNA so as to accelerate the formation of blood vessels and promote cell proliferation, and by influencing the expression of apoptosis-related factors-bcl-2/bax and reducing apoptosis so as to heal wound tissue. The paper expounded the healing mechanism of Yukuigao Ointment in the treatment of skin ulcer from the aspects of molecule and protein level. The study enriches the experimental methods and ideas in treating ulcer. |
