| Objective:to Compare the effect of relieving cough and resolving phlegm among different species of Chuan Fritillaria from the degree of Pharmacodynamics and clinical factors, and to find the specie of Chuan Fritillaria with the most significant effect of relieving cough and resolving phlegm, thus to provide the objective evidence for industrialization of authentic ingredients.Methods:(1) 70 Healthy guinea pigs had been randomly divided to 7groups:the Control group, the coughing model group, the Positive control group, the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria cirrhosa D. Don group and the Fritillaria przewalskii Maxim group. Continuously gave them herbs for 3 days.60s Citrate had been inhalated about 30 minutes after the last time of delivery. The Latency of cough and frequency of cough within 5 minutes were recorded.(2) 60 Healthy mice had been randomly divided to 6 groups:the control group, the positive control group, the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria cirrhosa D. Don group and the Fritillaria przewalskii Maxim group. Continuously gave them herbs for 3 days.30 minutes after the last time of delivery,0.5ml of 0.5% Phenol red solution had been intraperitoneally injected for each mouse. The mice were killed after 30 minutes. Mice's trachea had been separated and douched; 0.1ml of 5% NaHCO3 was added. After centrifuge manipulation, the supernatant had been divided. Enzyme labeling had been applied to measure the OD and eliminating phlegm index.(3) The methods of tratified randomization, double blinding and multi-center trial were applied in the clinical research.216 cases of acute bronchitis (cough due to heat phlegm) from 4 hospitals had been divided to 2 groups(the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group). Each hospital had to finish 54 cases (27 cases for Fritillaria unibracteata Hsiao et K.C.Hsia group and 27 cases forFritillaria delavayi Ftanch group). After 5 days of treatments, the therapeutic index and Safety indicators were observed such as cough score, marks of difficult phlegm, score for heat phlegm syndrome, the side effect in clinic and so on.Results:(1) The differences in cough latency of guinea pigs due to citrate:the model group had the shortest cough latency while the codeine group had the longest one. Within 4 species of Chuan Fritillaria, the cough latency in the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group and the Fritillaria cirrhosa D. Don group became shorter successively. The differences between model group and the codeine group, the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group were statistically significant (p<0.05). The differences between the codeine group and the Fritillaria unibracteata Hsiao et K.C.Hsia group,the Fritillaria delavayi Ftanch group,the Fritillaria przewalskii Maxim group and the Fritillaria cirrhosa D. Don group were statistically significant (P<0.05). The difference between the Fritillaria cirrhosa D. Don group and the Fritillaria unibracteata Hsiao et K.C.Hsia group was statistically significant (P<0.05). The difference between the Fritillaria cirrhosa D. Don groupand the Fritillaria delavayi Ftanch group was statistically significant (P<0.05).(2) The comparison in cough frequency of guinea pigs due to citrate within 5 minutes:the modeling group had the most frequency of cough while the codeine group had the least one. Within 4 species of chuan fritillaria, the cough frequency in the Fritillaria przewalskii Maxim group, the Fritillaria cirrhosa D. Don group, the Fritillaria unibracteata Hsiao et K.C.Hsia group and the Fritillaria delavayi Ftanch group became less in turn. The differences between the model group and the codeine group, the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group were statistically significant (P<0.05). The differences between the codeine group and the Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group as well as the Fritillaria cirrhosa D. Don group were statistically significant (P<0.05).(3)The OD value of Phenol red excretion for mice's trachea:the OD value in control group was the least one. The mcosolvan goup had the maximum OD value. Within 4 species of Chuan Fritillaria, the OD value in The Fritillaria unibracteata Hsiao et K.C.Hsia group, the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group, the Fritillaria cirrhosa D. Don group became smaller and smaller. The differences between control group and the mcosolvan goup, the Fritillaria unibracteata Hsiao et K.C.Hsia group as well as the Fritillaria delavayi Ftanch group were statistically significant (P<0.05). The differences between the mcosolvan goup and the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group as well as the Fritillaria cirrhosa D. Don group were statistically significant (P<0.05) The differences between the Fritillaria unibracteata Hsiao et K.C.Hsia group and the Fritillaria delavayi Ftanch group, the Fritillaria przewalskii Maxim group as well as the Fritillaria cirrhosa D. Don group were statistically significant (P<0.05).(4) The cough symptom was released in both groups, the difference was statistically significant (P<0.05). For stopping cough, the recovery rate in the Fritillaria unibracteata Hsiao et K.C.Hsia group was better than that of the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05). For the markedly effective rate and the total effective rate, the therapeutic effect of the Fritillaria unibracteata Hsiao et K.C.Hsia group was little better than the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05).(5) The difficult phlegm was released in both groups, the difference before and after treatment was statistically significant (P<0.05). For resolving phlegm, the recovery rate in the Fritillaria unibracteata Hsiao et K.C.Hsia group was better than that of the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05). The result of FAS was consistent with that of PP. For the markedly effective rate and the total effective rate of resolving phlegm, the therapeutic effect of the Fritillaria unibracteata Hsiao et K.C.Hsia group was little better than the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05).(6) The symptom scores of TCM were released in both groups, the difference before and after treatment was statistically significant (P<0.05). For therapeutic effect for heat phlegm syndrome, the recovery rate in the Fritillaria unibracteata Hsiao et K.C.Hsia group was a little better than that of the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05). For the markedly effective rate and the total effective rate of heat phlegm syndrome, the therapeutic effect of the Fritillaria unibracteata Hsiao et K.C.Hsia group was little better than the Fritillaria delavayi Ftanch group. But the difference had no statistically meaning (P>0.05).Conclusion:(1) Four species of Chuan Fritillaria may have some differences in stopping cough and resolving phlegm. Compared with Fritillaria przewalskii Maxim and Fritillaria cirrhosa D. Don, the Fritillaria unibracteata Hsiao et K.C.Hsia and the Fritillaria delavayi Ftanch may have the better effect of relieving cough and resolving phlegm. (2) Both of the Fritillaria unibracteata Hsiao et K.C.Hsia and the Fritillaria delavayi Ftanch had marked effect of relieving cough, the difference had no statistically meaning. The Fritillaria unibracteata Hsiao et K.C.Hsia may have a better effect in relieving cough.(3) Both of the Fritillaria unibracteata Hsiao et K.C.Hsia and the Fritillaria delavayi Ftanch had marked effect of resolving phlegm, the difference had no statistically meaning. The Fritillaria unibracteata Hsiao et K.C.Hsia may have a better effect of resolving phlegm than the Fritillaria delavayi Ftanch.(4) Either Fritillaria unibracteata Hsiao et K.C.Hsia or Fritillaria delavayi Ftanch can be separately used to treat "heat phlegm cough". Both of them can release the symptoms of chest tightness, dry throat, yellow urine and dry movement. The difference in therapeutic effect had no statistically meaning. The Fritillaria unibracteata Hsiao et K.C.Hsia may have a better effect for cough of heat phlegm syndrome than the Fritillaria delavayi Ftanch.To sum up, the Fritillaria unibracteata Hsiao et K.C.Hsia may have a better effect of relieving cough and resolving phlegm compared with Fritillaria delavayi Ftanch, Fritillaria cirrhosa D. Don and Fritillaria przewalskii Maxim. |
