PartⅠTHE EXPRESSION OF HYPOXIA INDUCIBLE FACTOR-1αAND MIGRATION INHIBITORY FACTOR IN ENDOMETRIOSISObjective:To investigate the expression of migration inhibitory factor and hypoxia inducible factor-1αin the eutopic and ectopic endometria of the endometriosis and in the normal endometria.To assess the association between migration inhibitory factor and hypoxia inducible factor expression and the endometriosis as well as the relation between the expression and the phases of menstrul cycle.Methods:15 cases of normal endometrial specimens,15 eutopic endometria of the endometriosis and 40 cases of endometriotic lesions were analyzed by RT-PCR,IHC, and Western blot for the detection of migration inhibitory factor and hypoxia inducible factor.Results:Both the eutopic and ectopic endometria of the endometriosis and the normal endometria expressed migration inhibitory factor and hypoxia inducible factor. The lengh of the amplified fragments was 159bp for HIF-1αand 127bp for MIF.The level of HIF-1αmRNA in the endometriotic lesions was obviousy higher than those in the eutopic endometria of the endometriosis as well as the normal ones,and the difference was statistically significant(P<0.05).Further more,the level of HIF-1αprotein in the endometriotic lesions was also obviousy higher than those in the eutopic endometria of the endometriosis as well as the normal ones,and the difference was statistically significant(P<0.05).When compared the levels of MIF among the same groups,it was easy to find that MIF was higher in the endometriotic group than the other groups both at the mRNA level and the protein level,and the difference was statistically significant(P<0.05).In addtion,there were significant differences of MIF between the eutopic endometria of the endometriosis and the normal endometria, and between the proloferative phase and the secretory phase.Conclusion:The levels of hypoxia inducible factor and migration inhibitory factor were highest in the endometriotic group compared to the eutopic endometrial group and the normal endometrial group.The levels of MIF in the eutopic endometria were higher in the proliferative phase,compared to those in the secretory phase.But there was no dramatic difference of the levels of HIF-1αin the eutopic endometria between the proliferative phase and the secretory phase.It seems that the two genes played a role in the development of endometriosis. PartⅡTHE RELATIONSHIP BETWEEN THE EXPRESSION OF HYPOXIA INDUCIBLE FACTOR-1αAND MAGROPHAGE MIGRATION INHIBITORY FACTOR AND THE CLINICAL STAGES AS WELL AS THE INFERTILITY IN HUMAN ENDOMETRIOSISObjective:To investigate the expression of hypoxia inducible factor-1αand migration inhibitory factor in the ectopic endometria of the endometriosis and in the normal endometria.To assess the association between migration inhibitory factor(or hypoxia inducible factor-1α) expression in the endometriosis and the status of infertility or the clinical stages.Methods:Fifteen cases of normal endometrial specimens and forty cases of endometriotic lesions were analyzed by Real time-PCR and Western-Blot for the detection of migration inhibitory factor and hypoxia inducible factor-1α.Patients were grouped by Revised American Fertility Society(rAFS) stages,seven patients atⅠorⅡstage,twenty-two atⅢstage,eleven atⅣstage.Nine of the forty endometriosis patients were infertile.The expression of hypoxia inducible factor-1αand migration inhibitory factor were analyzed according to the status of infertility or the clinical stages.Results:Both the ectopic endometria of the endometriosis and the normal endometria expressed hypoxia inducible factor-1αand migration inhibitory factor. The levels of HIF-1αmRNA in the ectopic endometriotic lesions of the 9 patients with infertility,the 31 fertile patients and normal endometria were 6.482±1.144,3.006± 1.129,0.450±0.115 respectively.The levels of MIFmRNA in the ectopic endometriotic lesions of the 9 patients with infertility,the 31 fertile patients and normal ensometria were8.261±2.757,3.399±0.754,0.134±0.070 respectively.The levels of HIF-1αprotein in the ectopic endometriotic lesions of the 9 patients with infertility,the 31 fertile patients and normal endometria were 1.034±0.182,0.751±0.177 and 0.348±0.116 respectively.The levels of MIF protein in the ectopic endometriotic lesions of the 9 patients with infertility,the 31 fertile patients and normal endometria were 1.397±0.315,0.861±0.197 and 0.448±0.154 respectively.The levels of HIF- 1αand MIF mRNA and protein were higher in the endometriosis patients with infertility than those in the patients without infertility.In the groupsⅠ/Ⅱ,ⅢandⅣ,the levels of HIF-1αmRNA were 3.758±2.022, 3.770±2.000,3.842±1.562 respectively;and the levels of HIF-1αprotein were 0.955±0.190,0.812±0.209,0.729±0.203 respectively.In the groupsⅠ/Ⅱ,ⅢandⅣ,the levels of MIF mRNA were 4.671±2.396,4.569±2.648,4.228±2.446 respectively; and the levels of MIF protein were 1.158±0.258,0.973±0.344,0.887±0.278 respectively.There were no obvious differences of the expression of MIF and HIF-1αamong the three groups.Conclusion:The levels of hypoxia inducible factor-1αand migration inhibitory factor were higher in the endometriotic lesions compared to the normal endometrial group.The levels of HIF-1αand MIF in the endometriosis patients with infertility were higher than those in the endometriosis patients without infertility.But the expression of HIF-1αand MIF seemed to be not related to the clinical stages.
