Considering the complexity of the material basis of Chinese herb, its extract is a complicated medical system that owns a lot of non-linear, multivariable and relative data characteristics, in which abundant biomedical information is stored. In the present study, in light of the technological system of "comparability evaluation of herbal medicine" in international medical field, it put forward:Chinese herb extract can be separated into basic active substance and accompanying substance. Basic active substance and the therapeutic characteristics of its preparations are completely or partly related, while accompanying substance can include additional and inactive by-products. Additional substance has the function of strengthening or weakening basic active substance. Inactive by-product has no pharmacological actions and its existence is often expected. By-product can change the physicochemical property of basic active substance, thus its biopharmaceutical parameter has been affected. In particular, it can affect the dissolving out of active substance from medical prescription or plant extract and its further absorption. How to make Chinese medical extractions acquire physicochemical property which can benefit the absorption of drugs is one of the important academic ideas of the present study. That is to say, physicochemical property of medical system has certain relations with internal absorption process of medicine.The present research project tries to study the influence of different accompanying substance to basic active substance in biopharmacy, explores and sets up characterizing method of physicochemical parameter of the composite of different accompanying substance and basic active substance, and introduce physicochemical parameter to function as the evaluation indicator of the Chinese medical extraction and during the extraction and separation process of Chinese drugs. PNS (Panax Notoginseng Saponins) which is composed of different accompanying substance is the research object. PNS extract is composed of two parts, which are basic active substance (total saponins) and accompanying substance (sugar, inorganic constituent, amino acid, microelement, sterols, and flavonoid, etc.).Firstly, the present study separated basic active substance (total saponins) by consulting the classical extraction and separation method and gradually reduced accompanying substance by using forward elimination method. It adopted ethanol to extract, macroporous resin to absorb, activate carbon to separate, and alumina column to purify. Hence, it set up preparation method of PNS of different accompanying substance; it also established determination method of HPLC, which is the main component of PNS of different accompanying substance, qualitative analysis of accompanying substance and finger-print research method of PNS of different accompanying substance, which has ensured the stable and even preparation of PNS of different accompanying substance.Secondly, it developed the measuring method of physicochemical parameters of viscosity, conductivity, turbidity and PH value, etc., which tended to be different. Judging from experimental data, there are certain laws to follow. Physicochemical changes before and after different extractions and purification may be the comprehensive superficial features of the process of removing the microcosmic by accompanying substance. It has provided evidence for finding out the physicochemical property of Chinese medical extractions which can benefit the absorption of drugs and seeking more appropriate evaluation system of material separation of Chinese medical effects.Then, it has studied the biopharmaceutical effects of accompanying substance on basic active substance. From the kinetics parameters of PNS Rl, ginsenoside Rgl, ginsenoside Rb1 of the PNS sample which is made of four different accompanying substances, it was observed that different accompanying substances have great impact on the absorption and elimination of PNS in the body. Besides, a common rule was observed, which is that accompanying with the increase of saponin purity in the sample and the decrease of accompanying substance, both Cmax of saponins in the body and AUC drop. The result has shown that accompanying substance in PNS can affect its bioavailability in the body.Finally, through multiple linear regression analysis, it discovered the relativity of physicochemical parameter (X) and the content of principal component of active substance (Y) by adopting neutral network and SVM (support vector machine) modeling discovery method. Moreover, it has done data mining of the relativity of physicochemical parameter (X) and pharmacokinetics parameter (Z). The result has shown that the fitting correlation coefficient of modeling neutral network and SVM of X and Y, X and Z are both above 0.95, which has provided reference for the feasibility of establishing evaluation system of physicochemical parameter of Chinese medicine. The present study made a primary research on the influence of accompanying substance on PNS in biopharmacy, the physicochemical parameters of PNS of different accompanying substances and the correlation between physicochemical parameters and accompanying substances, PNS bioavailability, its innovation consists in:I. Firstly it makes a preliminary study on the innovative idea that Chinese herb extract is composed of basic active substance and accompanying substance;â…¡. It establishes the determination method of the main physicochemical parameters of PNS of different accompanying substance;â…¢. It preliminarily explores the correlation between physicochemical parameters and accompanying substances, PNS bioavailability by adopting computer chemistry methods, such as multiple linear regression analysis, neutral network and SVM (support vector machine), and sets up the feasibility of establishing evaluation system of physicochemical parameter of Chinese medicine.
