The value orientation of informatization nowadays is to eliminate information-poor, decrease digital gap and make people in every corner of the world to enjoy the benefit of science progress. Spatial difference of regional informatization refers to the non-equal phenomenon among different regional systems. The research of spatial difference of regional informatization is a proposition reflecting scientific spirit and humanistic concern. As it is the theoretical entry point to research on space-dependence and regional principle of informatization on one hand, and it is the basis to make decision considering efficiency and fairness of informatization on the other hand.This paper absorbs the quintessence from Structural Theory of Space, New Economic Geography and Economics of Information and take the theoretical research and positive research systematically to the conception, estimation, framework, cause of formation, effects, regulation of the spatial difference of regional informatization and spatial model of informatization. This paper builds up the evaluation system of spatial difference of regional informatization and describes the general condition of spatial difference of regional informatization used the ways and means of multi-index comprehensive assessment, Analytic Hierarchy Process, Coefficient of Variation and Cluster analysis. The paper explains the reason for spatial non-equal development of regional informatization in the ways of factor analysis, Correlative analysis and regression model from to two angles of exogenous disturbance and internal originated demand. Taking the spatial effect, location choice and substitution of spatial function as the breakthrough point, the paper analyzes the influence of informatization to regional spatial structure. The paper appraises objectively the positive and negative effect causing by spatial difference of regional informatization , and points out the basic thinking to regulate the spatial difference of regional informatization. On the basis of the idea of overall coordinated development in the regional informatization, the paper builds up three model to develop regional informatization, namely taking center as the domination, urban and rural promoting each other and forward direction coupling. Taking Jiangsu Province as the sample, it gives three way to develop regional informatization as a whole, which are policy making, spatial arrangement and important fields.The research suggests: (1)The regional informatization has the features that the spatial differences, the graded differences and the function of two-sectors are all obvious, and the soft factors of informatization have the balanced spatial state. (2)The spatial differences are the results of operating law to organize, choose and cooperate with certain regional environment. (3)The spatial effect of informatization depends on centralization and decentralization of the regional system. (4)The spatial differences are premise to realize effective deploy of information resources, and also are the causalities of information power unbalance. To regulate the spatial differences, it is the right way to build up effective developing model of regional informatization.The value of this paper includes three aspects: first, it builds up an quantity evaluation system to appraise posture and cause of spatial differences of regional informatization; second, it explains the spatial centralization and decentralization of factors of regional informatization and the interrelation of informatization and region; third, it builds up the objective developing model of regional informatization, which accords with the regular pattern of spatial informatization and the needs of development of china.
