| Breast cancer is one of common malignant tumors in women. The treatment of breast cancer has usually included surgery, radiotherapy, Endocrine therapy, chemotherapy, immunotherapy, and traditional Chinese medicine therapy. In recent years, as radiotherapy, chemotherapy and surgical techniques continue to improve, the quality of life and prognosis of patients has been greatly improved. Chemotherapy is one of the most commonly used in cancer treatment, but chemotherapy alone is difficult to achieve a satisfactory therapeutic effect. It generally believed that early postoperative adjuvant chemotherapy should be applied, and the effect of combination chemotherapy superior to single agent chemotherapy. So to find a low toxicity and efficient prevention and treatment of breast cancer drugs is currently a hot research.Rhizoma Paridis is a traditional Chinese herbal medicine, it has been used widely for cancer treatment for a thousand years. Recent studies have shown that Rhizoma Paridis has significant antifebrile, alexipharmic, detumescent, demulcent, and hemostatic effects and is useful in the treatment of hepatopathy. Studies have also shown that steroid saponins are the main components in Rhizoma Paridis. In this study, Rhizoma Paridis saponins (RPS) was got by butanol-extraction after ethanol-extraction, and studied in vivo and in vitro on MCF-7 human breast cancer cell line inhibition and indepth study of its anticancer mechanism.When detecting mitochondrial membrane potential of MCF-7 cells, mitochondrial membrane potential gradually decreased with the RPS concentration increasing. It indicated that the cell apoptotic signal has been received, and the apoptosis process started. Apoptosis-related factors in MCF-7 cells were detected by Western Blot, it showed that the activation of PARP, capsase-3 and Caspase-9 levels increased with the RPS concentration, and the ratio of Bax/Bcl-2 increased significantly (P<0.05), cytosolic cytochrome C content are significantly increased compared with control group. The results showed that when RPS increased in the concentration, the cell started to mediate mitochondrial apoptosis pathway, including the reduction of mitochondrial membrane potential, increasing of Bax/Bcl-2 ratio, releasing of mitochondrial cytochrome C and activation of PARP and the Caspase cascade.After oral administrated tumor-bearing Nude mice models with the high and low doses of RPS for 14 days, the growth of tumor was inhibited significantly (P<0.05). The weight inhibition rate was 43.14% and 38.90%, and no significant toxicity of liver and kidney performed.Compared with the control group, the high and low doses of RPS treatment could increase the thymus index of tumor-bearing nude mice significantly (P <0.01), but the protection effect of RPS on spleen tissue was not significant.By detecting related cytokines in the serum of tumor-bearing nude mice, it’s showed that the levels of serum IFN-γand IL-2 were up-regulated, the level of serum IL-4 was down-regulated significantly (P<0.05), then the balance of Th1/Th2 shifted to Th1. Meanwhile, RPS treatment could also decrease the level of serum TNF-αsignificantly, which deprived the necessary conditions of growth and metastasis of tumor cells, and ultimately to inhibit the growth of tumor.RPS treatment can improve the tumor-bearing nude mice serum total antioxidant capacity (T-AOC) levels (P <0.01) and total superoxide dismutase (T-SOD) levels (P <0.05), and decreased serum MDA (MDA) levels (P <0.05) significantly. By analysing liver and kidney of tumor-bearing nude mice, it showed that T-AOC and T-SOD levels in tissues were increased significantly (P <0.05), but glutathione reductase (GR) levels were not significantly affected (P> 0.05).In this paper, Balb/c tumor-bearing nude mice and MCF-7 human breast cancer cell lines were acted as experimental material in vivo and in vitro. by analysing related proteins, cytokines, pathological structures and apoptosis thorough the methods of HE staining, ELISA, MTT method, flow cytometry and Western Blot techniques, et al, the effects and mechanisms of RPS anti-breast cancer was studied to provide a scientific basis and theoretical basis in cancer treatment. |
