Research on relationship between coronary heart disease and periodontitis about heat shock protein60and antibody in elderlyPurpose:To investigte heat shock protein60(HSP60), heat shock protein60IgG (HSP60IgG) and Porphyromonas gingivalis heat shock protein60IgG (P.gHSP60IgG) relationships between coronary heart disease and periodontitis,so as to understand the new risk factors of coronary heart disease with providing theoretical basis.Methods:Recruit a total of160veteran cadres in the Second Hospital of Tianjin Medical University in September to October2012. They were divided into four groups:the healthy control group, the group of coronary heart disease, the group of periodontitis, the group of coronary heart disease with periodontitis, each group contained40patients in average.The concentrations of HSP60, HSP60IgG, P.gHSP60IgG were determined by enzyme-linked immunosorbent assay (ELISA), and at the same time check the general biochemical markers such as liver and kidney function, blood lipids,and so on.Results:1.The concentrations of HSP60,HSP60IgG,P.gHSP60IgG,in the groups of coronary heart disease, periodontitis disease, coronary heart disease with periodontitis disease were significantly higher than the healthy control group (P<0.05). The patients of CHD with periodontitis disease had significantly higher HSP60, HSP60IgG,P.gHSP60IgG concentrations than another groups (P<0.05).The patients in the group of periodontitis disease had higher P.gHSP60IgG concentration than the group of CHD(P<0.05),and the concentrations of HSP60,HSP60IgG had no differences (P>0.05).2.The female in CHD group had higher HSP60concentration than the male(P<0.05), The female in the group of periodontitis disease had higher P.gHSP60IgG concentration than the male(P<0.05), The female in the group of CHD and periodontitis disease had higher HSP60IgG,P.gHSP60IgG concentration than the male(P<0.05).3.The HSP60,HSP60IgG, P.gHSP60IgG concentrations in all groups except the health control group tended to increase with age.The concentration of HSP60above80years old elderly in the three groups were significantly higher than the patients less than80years old (P<0.05),and the HSP60IgG, P.gHSP60IgG concentrations of the group of CHD and periodontitis disease above80years old were significantly higher than the patients less than80years old (P<0.05).The P.gHSP60IgG concentration of the group of periodontitis disease above80years old were higher,but there was no significance(P>0.05).4.The HSP60,HSP60IgG, P.gHSP60IgG concentrations in all groups tended to increase with BMI. The HSP60,HSP60IgG concentrations of BMI≥30in the group of CHD and periodontitis disease were significantly higher, compared to the another groups of which BMI were less than30(P<0.05).5.The concentrations of HSP60in smoking subgroup of CHD and periodontitis disease were significantly higher than the non-smoking subgroup (P<0.05).There was no significant smoking effect in other groups to the biological indicators(P>0.05).6.In addition to the healthy control group, the concentrations of HSP60, HSP60IgG in drinking subgroup were significantly higher than the non-drinking subgroup (P<0.05).7.The concentrations of TG,TCin the group of CHD and periodontitis disease were significantly lower than other groups,but the HSP60,HSP60IgG, P.gHSP60IgG concentrations were significantly higher(P<0.05).8.In the control of age,height,weigh,smoking,drinking,TG and other factors,the partial correlation analysis showed that HSP60and P.gHsp60IgG were positively correlated with human HSP60IgG(r=0.705,0.825,P<0.01).9.After the adjustment for traditional risk factors,such as age, gender, smoking, drinking,and so on,multivariate Logistic regression analysis showed that HSP60, P.gHsp60IgG and human HSP60IgG were respectively potential risk factors for coronary heart disease.Conclusions:HSP60,HSP60IgG and P.gHSP60IgG may play bridging roles between coronary heart disease and periodontitis, and the patients with periodontitis disease in the elderly may be more susceptible to coronary heart disease. In addition to the diagnoses of combination of ECG, clinical symptoms and coronary angiography,the determination of HSP60, HSP60IgG and P.gHSP60IgG concentrations may be instructive to assess whether the patients with periodontitis disease suffered from coronary heart disease or not.They may attribute to help prevent coronary heart disease by keeping oral health and prompt treatment of periodontitis disease in elderly. Purpose:To observe the changes of VE-cadherin,eNOS,Caspase-3protein expression and the tendency of cell proliferation and apoptosis after the intervention of human umbilical endothelial cells with porphyromonas gingivalis heat shock protein60by different concentrations and time points,and clarify the specific mechanisms of P.gHSP60inducing HUVEC dysfunction.Methods:P.gHSP60intervened HUVEC by the concentrations of10pg/mL,100pg/mL,1ng/mL at different time points such as2hours,6hours,12hours,24hours in order to clear the mechanisms of cell dysfunction,proliferation and apoptosis using the technology of Western bloting,PCR,MTT colorimetric assay,Annexin V-FITC/PI.Results:1. The protein expression had no differences at different time points when P.gHSP60intervene HUVEC by the concentrations of lOpg/mL.The protein expression of VE-cadherin, eNOS decreased after24hours intervention by the concentrations of100pg/mL, Caspase-3expression began to increase.When the intervention concentration of P.gHSP60increased to lng/mL,VE-cadherin, eNOS expression decreased after6hours, Caspase-3expression was opposite change.In short,the protein expression of Caspase-3enhanced by a concentration-time dependent way when P.gHSP60intervene HUVEC, but VE-cadherin and eNOS expression showed the opposite tendency.2.The VE-cadherin mRNA expression had no differences at different time points when P.gHSP60intervened HUVEC by the concentrations of lOpg/mL.The VE-cadherin mRNA expression decreased after12hours intervention by the concentrations of100pg/mL.When the intervention concentration of P.gHSP60increased to lng/mL,VE-cadherin mRNA expression decreased after6hours,and decreased by time dependent way.3.When P.gHSP60intervened HUVEC for24hours by the different concentrations,the ability of cell proliferation gradually reduced.4.When P.gHSP60intervened HUVEC for24hours by the different concentrations,the rate of early apotosis gradually increased.Conclusions:When P.gHSP60intervened HUVEC by a concentration-time dependent way,the protein expression of VE-cadherin, eNOS gradually decreased,and Caspase-3expression increased oppositely. When P.gHSP60intervened HUVEC for 24hours by the different concentrations,the ability of cell proliferation gradually reduced,even the rate of early apotosis gradually increased.From the point of molecular biology the study confirmed the mechanisms of atheroslerosis that the patients with periodontitis disease were prone to develop as a result of P.gHSP60.
