Tujia ethnic minority is one of the minorities owning more populous in China, has a population of over6million living mainly in Hunan, Hubei, Chongqing and Guizhou provinces. Tujia people accumulate abundant knowledge and experience of using Chinese traditional plant medicine through long-term daily work and curing disease and form its own nationality characteristic medical theory. To improve application of Tujia ethnic plant medicine, and discover new active lead compound, this study focus on two Tujia ethnic plant medicine, Schisandra propinqua and Pteris multifida Poir. and Sudachi peels using a variety of isolation and purification technique and spectrometric methods such as HR-MS、NMR、IR、and UV to find new compounds and active compounds.1. The plant family Schisandraceae, consisting of the genera Schisandra and Kadsura, contains several important medicinal plants. In this study, we report on the isolation of seven new lignans, eleven known lignans and two sesquiterpene. Compounds14-16and18were isolated from Schisandra propinqua for the first time. In the bioactivity assay, compound1,3,8,9and11revealed a significant immunosuppressive activity and compound11also exhibited significant anti-metastasis activity (IC50=10.30μg/mL).2. Pteris multifida Poir.(Pteridaceae) is a plant widely distributed in the southeast of China, and the whole pant has been used as traditional Chinese medicine and an antitumor and anti-inflammatory agent. Previous phytochemical investigation of the genus Pteris revealed C-20-nonoxygenated ent-kaurane diterpenoids and C14sesquiterpenoids (pterosins) with1-indanone skeletions. In this study, we isolated10compounds using advanced separation and purification technique and spectrometric methods and compounds28~30were isolated from this plant for the first time. In the bioactivity assay, compounds21~25stimulate glucose consumption of insulin-resistance HepG2cells significantly and compound21is the most promising compound. In the further investigation, compound21revealed significant activation of phosphorylation of AMPK and its substrate, ACC. And Compound C, a highly-selective AMPK inhibitor, attenuated the21-stimulated glucose consumption and phosphorylation of AMPK. These results indicate that AMPK is a key factor in21-induced glucose consumption.3. Citrus sudachi Hort. Ex Shirai (Rutaceae), called "sudachi", is an evergreen tree founded mainly in Tokushima Prefecture in Japan. Sudachi is a very well-known citrus fruit in the south part of Japan. Annual Production of the fruit is8000tons. Half is sold as an acidulant for seasoning, and the remaining half is used for juice. The residue of sudachi juice remains as food industrial waste. Previous research of Sudachi indicated it can significantly decrease blood glucose level and blood lipids level. In this study, Sudachi peal extraction was separated into different polar fractions and the activity of each fraction was assay by cell experiment in vitro. Hexane fraction significantly increased glucose uptake of C2C12cells. And hexane and ethyl acetate fractions enhanced the SIRT1activity of C2C12. Thus hexane and ethyl acetate fractions maybe the active fractions. In addition, the activity of34compounds isolated from Sudachi was assayed, compounds1,10and12significantly enhance SIRT1activity of C2C12without effect the protein level of SIRT1, indicating these compounds might regulating glucose/lipid metabolism and life span.
