| Objective:To investigate the effecet of Chongcaoyishen decoction on the renal interstitial fibrosis(RIF) induced by unilateral ureteral obstruction in rats, and to explore the mechanism.Method:The 200 male wistar rats were randomly divided into five groups:normal group, sham operation group, UUO group, Chongcaoyishen group, fosinopril group, each group had 40 rats. The rats were given daily intragastric administration from the first day after UUO surgery. Normal group, sham operation group and UUO group were given distilled water, while Chongcaoyishen group was given Chongcaoyishen decoction and fosinopril group was given fosinopril solution. Ten rats were randomly chosen to put into the metabolism cage and then executed the next day after the UUO surgery to collect the 24 hour urine to detect the quantitate of 24h proteinuria, serum to detect Scr and BUN,kidney to detect the pathological change and comparative proteomics test.Results:1.The status of rats in Chongcaoyishen decoction were ameliorative distinctly after UUO,and the wight and urine volume were higher than UUO group(P< 0.05).2. The biochemical detection results showed that the level of quantitate of 24h proteinuria, Scr and BUN in Chongcaoyishen group were lower than in UUO group(P< 0.05).3. The pathological detection results showed that rats in Chongcaoyishen group had less fibrosis area than UUO group(P< 0.05).4. The proteomics results showed that 6793 protein were identified in UUO and normal group, the distinct protein number was 1862, of which there were 875 up-regulation protein and 987 down-regulation protein.5. The bioinformatics analysis results showed that the distinct protein involved in variety biological process such as cellular process, metabolic process, biological regulation, response to stimulus, signaling, immune system process, biological adhesion, growth, etc, while 315 KEGG pathway were related, such as PI3K-Akt, focal adhesion, PPAR signaling pathway, MAPK signaling pathway, HIF-1 signaling pathway, TNF signaling pathway, Toll-like receptor signaling pathway, VEGF signaling pathway, Wnt signaling pathway, Notch signaling pathway, TGF-Psignaling pathway, Jak-STAT signaling pathway, etc.6. Western blot results showed that compared with UUO group the level of Wnt4 and β-catenin was lower in Chongcaoyishen group(P< 0.05).Conclusion:1. Chongcaoyishen decoction can ameliorative the status of rats and increase the weight and urine volume.2. Chongcaoyishen decoction can reduce the proteinuria and the level of Scr and BUN,and improve the renal function.3. Chongcaoyishen decoction can alleviate the renal pathological change,and shrink the fibrosis area.4. The mechanism of UUO involves a lot of signal pathway and cytokines.5. Chongcaoyishen decoction can play an anti-fibrosis effect, and the mechanism may due to suppress the Wnt/β-catenin signal pathway. |
