| Object: To investigate the effect and to explore the mechanism that GushangfuyuanDecoction(GSFYD)intervenes the traction distract periosteum of callus,within theperiosteum and the center of callus to the bFGF,TGF-β,bFGF-mRNA and TGF-β1mRNAby distraetion osteogenesis.Methods:The study selects72healthy rabbits (male and female) weights range1.5±0.3kg,divided into three groups randomly with each group has24rabbits,theobservation group,the control group and the model group,then the three groups dividedagain into four groups randomly with each group has6rabbits.The three groups were usedthe same model operation of bone defection of the tibia,after the operation they were giventhe equal dose by the stomach(the model group was used the Distilled water,the controlgroup was used the water Solution of Jieguqilipian and the observation group was used theGSFYD)At the first,second and third week and after pulling treatment one week, we tookthe Materials from the traction distract periosteum of callus,within the periosteum and thecenter of callus to do the Hybridization experiment on the bFGF,TGF-β,bFGF-mRNAand TGF-β1mRNA In situ and measure the the average of optical density to observe theexpressions.Results:Based on the experiment,we found that bFGF and bFGF-mRNAã€TGF-β1and TGF-β1mRNA can make the consistent expressions.after the first week,theobservation group and the control group were apparently better than the model group bycomparing the traction distract periosteum of callus expressions with the bFGF andbFGF-mRNA〠TGF-β1and TGF-β1mRNA (p<0.05).There are no significantdifferences among the observation group and the control group(p>0.05).By comparing within the periosteum and the center of callus expressions:There are no significantdifferences among the observation group,the control group and the model group(p>0.05).after the second week,by comparing the traction distract periosteum of callusexpressions with the bFGF and bFGF-mRNAã€TGF-β1and TGF-β1mRNA,the controlgroup was apparently better than the model group(p<0.05),the observation group wasapparently better than the control group(p<0.05).At the second treatment the observationgroup got the peak point and was more significantly than the model group(P<0.01).Bycomparing within the periosteum and the center of callus expressions:the observationgroup and the control group were apparently better than the model group(p<0.05).Thereare no significant differences among the observation group and the control group(p>0.05).after the third week,by Observing the traction distract periosteum of callus andwithin the periosteum expressions with the bFGF and bFGF-mRNAã€TGF-β1andTGF-β1mRNA:the observation group and the control group was apparently better than themodel group(p<0.05),There are no significant differences among the observation groupand the control group(p>0.05).the center of callus expressions:the control group wasapparently better than the model group(p<0.05),the observation group was apparentlybetter than the control group(p<0.05).At the third treatment the observation group got thepeak point and was more significantly than the model group(P<0.01).After stopped pullingthe callus one week,the traction distract periosteum of callus,within the periosteum and thecenter of callus to do the Hybridization experiment on the bFGF,TGF-β,bFGF-mRNAand TGF-β1mRNA:the observation group and the control group were apparently betterthan the model group(p<0.05).Conclusion: In the rabbit model of tibial leagthening,the GSFYD treatment with bonetransport can improved and Stimulated the expression levels of bFGF,TGF-β,bFGF-mRNAand TGF-β1mRNA vs.Control and model groups.It can farther boostthe bone formationand reduee the treatment time. |
