| Objective:Blast injury is a complex type of physical trauma resulting from director indirect exposure to an explosion. Blast lung injury of patients is strongly associatedwith the poor outcome, severity of illness and high incidence of mortality.Liquidventilation with perfluorocarbon can significantly improve the blood oxygen saturationlung compliance,ventilation perfusion ratio,alleviating inflammatory reaction of thelung in patients with acute lung injury. A549cells were selected as our researchobject.The effects and mechanism of perfluorocarbon on blast induced lung epithelialtype II cells injury, provides the theory basis for the clinical treatment of blast lunginjury.Methods:1.100Kpa shock wave induced A549cells injury, the cells were dividedinto two groups which were control group (C), blast injury group (B).Cell biology, cellmorphology and the mRNA of inflammatory factors were detected.2. PFC were used tomodulate blast-induced A459cell injury model, and the cells were divided into4groups:control group (C), blast group (B), blast+perfluorocarbon group (B+P), perfluorocarbongroup (P). Thecell morphology, cell biology, inflammatory cytokine and reactiveoxidative reaction were detected respectively.3. MAPK, Bcl-2, Bax, CleavedCaspase-3and NF-κB protein expression were detected by western blot and high-throughputstudies using LncRNA microarray.Results:1. Cell biology and cell morphology ofA549cells in blast group was poorcompared with control group.The cell proliferation and migration was inhibited by theblast wave and the inflammatory factors such as IL-1β, IL-6, TNF-α mRNA weresignificantly increased compared with control group.2.PFC intervention can attenuatethe suppression of the migration and proliferation of A549cells induced by blast wave; PFC can down regulate the increasing of the inflammatory cytokines and oxidativestress factors induced by blast wave;PFC can reduce cell apoptosis induced by shockwave.3.PFC can attenuate the protein expression of MAPK and NF-κB in blast waveinduced A549cells, with no significant effect on untreated A549cells.Conclusion:1.PFC has protective effects on the shock wave induced damage inA549cells.2. PFC can reduce inflammation and oxidative stress reaction via downregulate MAPK and NF-κB signaling pathway.3.PFC can reduce apoptosisof A549cells via down regulate Bax, Caspase-3, up regulation of Bcl-2protein... |
