| Objective: Yiqibushenhuoxue prescription is empirical formula to treat disuseosteoporosis. The paper studies the pharmacology effects by animal experiment, studiesthe mechanism of the Yiqibushenhuoxue treating disuse osteoporosis, and providethe scientific basis and the effective prescription for the treatment of bone loss resultingfrom long-term brake.Methods: Experimental study: the disuse osteoporosis model was established by theoperation of braking method with serum pharmacology methods, technology, combinedwith experiments in vivo and in vitro, combined with the cell culture in vitro, using fourtetrazolium method, EIA method, ELISA method, wheat germ agglutination method,discusses in detail the Yiqibushenhuoxue prescription in the treatment of waste with themechanism of osteoporosis. theoretical research: in this study, under the guidance ofChinese medicine theory, according to literature to discuss therapeutic effect ofYiqihuoxuebushenhuoxue prescription on disuse osteoporosis, to illuminate the Principleof formula and compatibility significance.Results: Through the experiment we found that Yiqibushenhuoxue prescription cansignificantly improve the disuse bone metabolism in osteoporosis rats.It can significantlyincrease bone mineral density (BMD) and bone mineral content (BMC).It can increase thewet weight and dry weight bone effectively.It can improve the structure and function of ratfemur and the expression of transforming growth factor TGF and the number and thicknessof trabecular bone.It can decrease the content of serum alkaline phosphatase (ALP), bonespecific alkaline phosphatase (BAP), urinary pyridinoline (PYD) and deoxypyridinoline(DPD). And it can increase the content of blood Ca, osteocalcin (BGP), bonemorphogenetic protein-2(BMP-2)ã€tartrate resistant acid phosphatase (TRAP) obviously. It can decrease the levels of tumor necrosis factor alpha (TNF-α), interleukin-4(IL-4),interleukin-6(IL-6) and increase the content of interleukin-10(IL-10).It can improvebone marrow mesenchymal stem cells and osteoclast proliferation and effectively increasethe bone alkaline phosphatase (BALP) activity. It can promote bone marrow mesenchymalstem cells into osteoblasts and effect the bone metabolism which is helpful for treatment ofdisuse Osteoporosis. While it can significantly promote the proliferation of osteoclast andreduce the number of resorption pits and improve the bone formind and reduce boneresorption to a certain extent.Conclusion: Yiqibushenhuoxue prescription can effectively reduce bone mass loss ofthe disuse osteoporosis model rats, inhibit bone resorption for braking, enhance boneformation. It provides new method and formulas for the clinical treatment of disuseosteoporosis. |
